Vitamin E Derivative Alpha-Tocotrienol Failed to Show Neuroprotective Effects after Embolic Stroke in Rats
Authors
Abstract:
Objective(s) Previous studies have demonstrated that pretreatment with alpha-tocotrienol (a-TCT) can reduce ischemic damage in mice following middle cerebral artery (MCA) occlusion. It is also reported to decrease stroke- dependent brain tissue damage in 12-Lox-deficient mice and spontaneously hypertensive rats. In the present study, the neuroprotective effects of a-TCT and rosiglitazone (RGZ) at 3 hr after cerebral ischemia were investigated. Materials and Methods Stroke was induced by embolizing a preformed clot into the MCA. Rats were assigned to vehicle, a-TCT (1 or 10 mg/kg), RGZ and sham-operation. Results Compared to the control group, only RGZ decreased infarct volume (P

full textAlpha-tocotrienol is the most abundant tocotrienol isomer circulated in plasma and lipoproteins after postprandial tocotrienol-rich vitamin E supplementation
BACKGROUND Tocotrienols (T3) and tocopherols (T), both members of the natural vitamin E family have unique biological functions in humans. T3 are detected in circulating human plasma and lipoproteins, although at concentrations significantly lower than α-tocopherol (α-T). T3, especially α-T3 is known to be neuropotective at nanomolar concentrations and this study evaluated the postprandial fate...
full textThe neuroprotective effects of Tao-Ren-Cheng-Qi Tang against embolic stroke in rats
BACKGROUND Combinations of the traditional Chinese and Western medicines have been used to treat numerous diseases throughout the world, and there is a growing body of evidence showing that some of the herbs used in traditional Chinese medicine elicit significant pharmacological effects. The aim of this study was to demonstrate the neuroprotective effects of Tao-Ren-Cheng-Qi Tang (TRCQT) in com...
full textTissue distribution of vitamin E metabolites in rats after oral administration of tocopherol or tocotrienol.
We previously found that 2,7,8-trimethyl-2(2'-carboxyethyl)-6-hydroxychroman (γCEHC), a metabolite of the vitamin E isoforms γ-tocopherol or γ-tocotrienol, accumulated in the rat small intestine. The aim of this study was to evaluate tissue distribution of vitamin E metabolites. A single dose of α-tocopherol, γ-tocopherol or a tocotrienol mixture containing α- and γ-tocotrienol was orally admin...
full textEmbolic stroke in aged rats.
BACKGROUND AND PURPOSE Although stroke is a disorder associated with aging, experimental studies of stroke are conducted in young adult (2-4-month-old) animals (rat life span, 27-29 months). To determine whether histopathologic changes caused by cerebral infarction would be altered in aged animals, we produced embolic cerebral infarction in 17 aged (23-24-month-old) and 16 young (2-4-month-old)...
full textThe neuroprotective effects of tocotrienol rich fraction and alpha tocopherol against glutamate injury in astrocytes.
Tocotrienol rich fraction (TRF) is an extract of palm oil, which consists of 25% alpha tocopherol (α-TCP) and 75% tocotrienols. TRF has been shown to possess potent antioxidant, anti-inflammatory, anticancer, neuroprotection, and cholesterol lowering activities. Glutamate is the main excitatory amino acid neurotransmitter in the central nervous system of mammalian, which can be excitotoxic, and...
