abstract
introduction demyelination is a common lesion of spinal cord injury; cell therapy is an approach for replacing the lost oligodendrocytes. objective in this study, bone marrow stromal cells (bmscs) have been transdifferentiated into oligodendrocyte-like cells (olcs) and used in cytotherapy of contused spinal cords in rats. materials and methods the bmscs were collected from rat long bones, cultured and characterized by different markers, then pre-induced with dimethyl sulfoxide followed by retinoic acid. then, the pre-induced cells were induced with a combination of basic fibroblast growth factor (bfgf), platelet derived growth factor (pdgf), and heregulin (hrg) followed by triiodothyronine (t3). the olcs were transplanted in the contused spinal cords of the rats combined with undifferentiated bmscs. specific markers were used to characterize the cells by immunohistochemistry and rt-pcr. results the bmscs showed typical immunoreactivity to the markers and the olcs were immunostained with specific markers. there was an improvement in bbb test with reduction in the cavitation in the contused rats treated with olcs combined with bmscs. conclusion the transplanted cells were detected in the contused spinal cord. the combination of the transdifferentiated bmscs into olcs with the undifferentiated bmscs improved the contused spinal cord.

Improvement of spinal contusion model by cotransplanting bone marrow stromal cells and induced BMSCs into oligodendrocytes-like cells.
BACKGROUND Demyelination is a common lesion in spinal cord injury, cell therapy is one of the approaches for replacing the lost oligodendrocytes. In this study, bone marrow stromal cells (BMSCs) have been transdifferentiated into oligodendrocyte-like cells (OLCs) and used in cytotherapy of contused spinal cords in rats. METHODS The BMSCs were collected from the rat long bones, and cultured an...
full textInduction of Bone Marrow Stromal Cells into Cholinergic-Like Cells by Nerve Growth Factor
Background: Bone marrow stromal cells (BMSC) are used as a source for cell therapy in different model for neurological disorder such as stroke and spinal cord injury. However, the transdifferentiation of BMSC into cholinergic phenotype requires more investigation. Methods: BMSC were isolated from adult rats, pre-induced with β-mercaptoethanol (BME) and followed by nerve growth factor (NGF) indu...
full textA New Multistep Induction Protocol for the Transdifferentiation of Bone marrow Stromal Stem Cells into GABAergic Neuron-Like Cells
Background: Bone marrow stromal stem cells (BMSC) are appropriate source of multipotent stem cells that are ideally suited for use in various cell-based therapies. It can be differentiated into neuronal-like cells under appropriate conditions. This study examined the effectiveness of co-stimulation of creatine and retinoic acid in increasing the differentiation of BMSC into GABAergic neuron-lik...

full textA Study on Transdifferentiation of Bone Marrow Stromal Cells into Neuronal and Glial-Like Cells In Vitro by Different Inducers
Introduction: There are some evidences to suggest that bone marrow stromal cells (BMSCs) not only differentiate into mesodermal cells, but also adopt the fate of endodermal and ectodermal cell types. BMSCs can be a valuable cell source as an autograft for clinical application involving regeneration of the central nervous system. Bone marrow stromal cells can be expanded rapidly in vitro and can...
full textTransplanting P75-Suppressed Bone Marrow Stromal Cells Promotes Functional Behavior in a Rat Model of Spinal Cord Injury
Background: Bone marrow stromal cells (BMSC) have been successfully employed for movement deficit recovery in spinal cord injury (SCI) rat models. One of the unsettled problems in cell transplantation is the relative high proportion of cell death, specifically after neural differentiation. According to our previous studies, p75 receptor, known as the death receptor, is only expressed in BMSC in...
