Blooms of cyanobacteria are frequent in Brazilian water reservoirs used for drinking water. The warning the presence potential toxin-producing is typically based on time-consuming microscopy, rather than specific molecular detection toxic genes cyanobacteria. In this study, we developed a quantitative PCR assay producing neurotoxin saxitoxin (STX). targets sxtA gene sxt cluster. Potential and d...
Carotenogenesis of Staphylococcus aureus: New insights and impact on membrane biophysical properties
Staphyloxanthin (STX) is a saccharolipid derived from carotenoid in Staphylococcus aureus involved oxidative-stress tolerance and antimicrobial peptide resistance. STX influences the biophysical properties of bacterial membrane has been associated to formation lipid domains regulation methicillin-resistance. In this work, targeted metabolomics characterization study was carried out investigate ...
Shigella dysenteriae are significant agents of bacillary dysentery, accounting for a considerable number illnesses with high morbidity worldwide. The Shiga toxin (Stx) encoded by defective prophage is the key virulence factor S. type 1 (SD1) strains. Here we present full genome sequence an SD1 strain HNCMB 20080 isolated in 1954, compare it to other sequenced genomes, and assess diversity Stx-p...
Specificity of PCR and serological assays in the detection of Escherichia coli Shiga toxin subtypes.
Specificity analysis for stx or Stx subtypes in Escherichia coli showed that the PCR assays we tested did not detect stx(1d) and stx(2f), and some also missed stx(2b) and stx(2g). Most of the serological assays examined did not detect Stx2c, Stx2e, Stx2f, and Stx2g, and some strain-to-strain variation in reactivity was observed for Stx2b.
This study reports the phenotypic and genotypic characterization of 144 Shiga toxin-producing Escherichia coli (STEC) strains isolated from urban sewage and animal wastewaters using a Shiga toxin 2 gene variant (stx(2))-specific DNA colony hybridization method. All the strains were classified as E. coli and belonged to 34 different serotypes, some of which had not been previously reported to ca...
Stx2d is a recently described Shiga toxin whose cytotoxicity is activated 10- to 1000-fold by the elastase present in mouse or human intestinal mucus. We examined Shiga toxigenic Escherichia coli (STEC) strains isolated from food and livestock sources for the presence of activatable stx(2d). The stx(2) operons of STEC were first analyzed by PCR-restriction fragment length polymorphism (RFLP) an...
The potencies of saxitoxin (STX) and of five structurally related toxins were determined by their ability to block impulses at equilibrium in frog sciatic nerve. The order of potency, with values relative to STX potency in parentheses, was: neo-STX (4.5) greater than gonyautoxin (GTX) III (1.4) greater than STX (1.0) greater than GTXII (0.22) greater than 12 alpha-dihydroSTX (0.050) greater tha...
Phages 933W, BAA2326, 434, and λ are evolutionarily-related temperate lambdoid phages that infect Escherichia coli. Although these are highly-similar phages, BAA2326 and 933W naturally encode Shiga toxin 2 (Stx⁺), but phage 434 and λ do not (Stx(-)). Previous reports suggest that the 933W Stx⁺ prophage forms less stable lysogens in E. coli than does the Stx(-) prophages λ, P22, and 434. The hig...
Previously, we have shown that chickens immunized with Shiga toxin (Stx) produce Stx-neutralizing egg yolk immunoglobulin Y (IgY) antibody. The anti-Stx-1 IgY and anti-Stx-2 IgY exert their neutralizing activity through their antibody activity against the B subunit of the toxin but not the A subunit. In the present study, chickens were immunized with recombinant Stx-1 B subunit (rStx-1B) and re...
