We describe a method for calculating the absorptivity (in terms of substrate consumed) of the colored solution obtained when o-dianisidine dihydrochloride is oxidized by ceruloplasmin. By oxidizing o-dianisidine dihydrochloride with known amounts of hydrogen peroxide we could determine that the molar reacting ratio of o-dianisidine to hydrogen peroxide is 2:1. So calculated, absorptivity is 9.6...

A simple, rapid, heat-stable (70 degrees C) acid-phosphatase test for differentiation of mycobacteria was developed with the alpha-naphthylphosphoric acid-diazonium o-dianisidine color reagent.

Directed evolution methods were developed for Cu-containing nitrite reductase (NiR) from Alcaligenes faecalis S-6. The PCR cloning strategy allows for the efficient production of libraries of 100 000 clones by a modification of a megaprimer-based whole-plasmid synthesis reaction. The high-throughput screen includes colony lift onto a nylon membrane and subsequent lysis of NiR-expressing colonie...

Laccase production by solid state fermentation (SSF) using an indigenously isolated litter dwelling fungus Fusarium incarnatum LD-3 was optimized. Fourteen medium components were screened by the initial screening method of Plackett-Burman. Each of the components was screened on the basis of 'p' (probability value) which was above 95% confidence level. Ortho-dianisidine, thiamine HCl and CuSO(4)...

We have developed a new method for determination of serum ceruloplasmin from its oxidase activity, in which o-dianisidine dihydrochioride is used as substrate. o-Dianisidine dihydrochloride is more stable than is the more widely used p-phenylenediamine substrate, and forms a stable product that is measured at 540 nm. Correlation was good between results of this method and those obtained with bo...

An automated method for the enzymatic determination of ceruloplasmin with o-dianisidine dihydrochloride as substrate is described. The method enables the measurement of 30 samples per hour with a coefficient of variation (day-to-day) of 2.8%. Results correlate well (r equals 0.99 with those obtained by the corresponding manual method

mercury is one of the three major environmental metal poisons, and mercuric chloride is a highly reactive compound which can harm cells by a variety of mechanisms including direct interaction with sulphydryl groups of proteins and enzymes, therefore affecting the enzymatic activity. this study focused on the effect of hg++ on horseradish peroxidase (donor: hydrogen peroxide oxidoreductase, ec 1...

THE DETERMINATION of glucose by the glucose oxidase-peroxidase system has been simplified by Hill and Kessler (2) with the use of an automated system first described by Skeggs (1). Wincy and Marks (3) have also developed a method using o-tolidine in the automated system. These procedures require a large quantity of sample and use of a dialyzer to separate the protein before color development. T...
