To study the distribution and localization of oil-soluble arteether in experimental mice, we injected C14-labelled arteether (20 microCi/kg body weight) intramuscularly and measured radioactivity in the blood, kidney, and liver. The labelled arteether distributed and localized more to the kidney (819,180.4 +/- 34,134 dpm/cm3) than the liver (288,628.9 +/- 54,954 dpm/cm3) 4 hours post-injection....
β-Arteether, an effective artemisinin derivative, is used in the treatment of malaria but available only as an intramuscular injection. The objective of this work was to develop lipid-based formulations for oral administration of β-arteether. Self-emulsifying drug delivery systems (SEDDSs) of low cost and with accessible excipients (groundnut or sesame oil, Maisine 35-1, Tween 80 or Cremophor E...
BACKGROUND & OBJECTIVES Morbidity and mortality due to falciparum malaria are increasing in many tropical areas. The situation is further complicated by drug resistant malaria. The present study was undertaken to evaluate the efficacy of arteether on acute chloroquine resistant Plasmodium falciparum malaria in eastern coalfield area of Asansol. METHODS A total of 30 patients with chloroquine ...
The purpose of the present work is to improve the antimalarial activity of arteether through enhancing its solubility subsequently bioavailability by incorporating the drug into the cyclodextrins cavity. The effect of hydrophilic polyvinyl propylene (PVP) polymer on the complexation and solubilizing efficiencies of cyclodextrins (CDs) is also elucidated. Inclusion of arteether molecule in solid...
The control of malaria has been complicated by the increasing resistance of malarial parasites to multiple drugs. However, artemisinin-based drugs offer hope in the fight against drug-resistant parasites. The mode of action of these drugs remains unclear, but evidence suggests a role for free radicals in their mechanism of action. In this study, we examined the relationship between the intracel...
Arteether™ is among the recent drugs that are used to combat chloroquine-resistant malarial parasites. This study examined the effects of arteether™ on enzyme biomarkers of the liver, serum protein concentrations, and liver morphology. Twenty (20) adult albino Wistar rats weighing 200 - 250 g were randomly divided into four groups (A, B, C and D) of five animals each, and used in this study. Gr...
The study evaluates in vivo antimalarial activity of arteether and garlic pearl oil combination in Plasmodium berghei-infected mouse model of malaria. 72 h (Day 3) post infection, at 2-4% parasitemia, mice were treated with single dose intramuscular injection of α-β arteether, at 750 μg, in combination with three 100 μL oral doses of garlic pearl oil on Day 3, Day 4 and Day 5. Following the tre...
Malaria is one of the world 's most devastating human infectious disease and, In India, it is endemic with the mortality rate steadil y increasing. As a result the search for the development of new and more effective chemotherapeutic agents to control this life threatening disease is going on at several centres. In this context, artemisinin isolated from the Chinese herb qinghao (Artemisia annu...
Artemisinin and curcumin show an additive interaction in killing Plasmodium falciparum in culture. In vivo, 3 oral doses of curcumin following a single injection of alpha,beta-arteether to Plasmodium berghei-infected mice are able to prevent recrudescence due to alpha,beta-arteether monotherapy and ensure almost 100% survival of the animals.
