multiple gene silencing is being required to target and tangle metabolic pathways in eukaryotes and researchers have to develop a subtle method for construction of rna interference (rnai) cassettes. although, several vectors have been developed due to different screening and cloning strategies but still some potential limitations remain to be dissolved. here, we worked out a simple cloning stra...

background: electroporation is a valuable tool for small interfering rna (sirna) delivery into cells because it efficiently transforms a wide variety of cell types. since electroporation condition for each cell type must be determined experimentally, this study presents an optimal electroporation strategy to reproducibly and efficiently transfect mda-mb 468 human breast cancer cell with sirna. ...

More antibody validation protocols would identify non-specific reagents - a major source of irreproducible research - with the inclusion of negative controls. This article presents an overview of one such method: siRNA knockdown. The authors outline a general protocol, the knockdown mechanism, and tips for evaluating knockdown experiments.

In this study, we used solid phase synthesis to prepare three kinds of peptides and then formulated their peptide-siRNA complexes and peptide-siRNA conjugates. Both the complexation and conjugation systems were nontoxic and allowed the delivery of siRNA into the cytoplasm without the need for any transfection agents and with subsequent inhibition of gene expression.

A systematic structure-activity relationship study of 4'-thioribose containing small interfering RNAs (siRNAs) has led to the identification of highly potent and stable antisense constructs. To enable this optimization effort for both in vitro and in vivo applications, we have significantly improved the yields of 4'-thioribonucleosides by using a chirally pure (R)-sulfoxide precursor. siRNA dup...

Both siRNA and antisense oligodeoxynucleotides (ODNs) inhibit the expression of a complementary gene. In this study, fundamental differences in the considerations for RNA interference and antisense ODNs are reported. In siRNA and antisense ODN databases, positive correlations are observed between the cost to open the mRNA target self-structure and the stability of the duplex to be formed, meani...
