Valproic acid can be determined by gas-chromatography as the free acid after its extraction (1 ) or derivatization to form methyl, butyl, or phenacyl estars (2-4), the latter methods being more complicated for routine analyses. Other procedures, such as enzyme immunoassay (Syva Co., Palo Alto, CA 94304) or fluoroimmunoassay (Miles Inc., Elkhart, IN 46515), which do not require serum extraction,...
A 42-year-old man with human immunodeficiency virus (HIV) infection and a history of complex partial seizures developed severe anemia after the addition of valproic acid to his stable antiretroviral regimen of zidovudine, lamivudine, and abacavir. The inhibition of zidovudine glucuronidation by valproic acid and the resultant zidovudine hematologic toxicity is the proposed mechanism of the inte...
BACKGROUND Epigenetic programming, dynamically regulated by histone acetylation, is a key mechanism regulating cell proliferation and survival. Little is known about the contribution of histone deacetylase (HDAC) activity to the development of pulmonary arterial hypertension, a condition characterized by profound structural remodeling of pulmonary arteries and arterioles. METHODS AND RESULTS ...
INTRODUCTION Valproic acid is an effective first line drug for the treatment of epilepsy. Hepatotoxicity is a rare and potentially fatal adverse reaction for this medicine. OBJECTIVE Firstly to characterise valproic acid reports on children with fatal outcome and secondly to determine reporting over time of hepatotoxicity with fatal outcome. METHODS Individual case safety reports (ICSRs) fo...
A girl aged 1 year died of acute haemorrhagic pancreatitis while taking sodium valproate. Necropsy showed widespread vascular disease that may have contributed to the onset of pancreatitis. Previous reports of pancreatitis in children receiving valproic acid are reviewed and although the association is rare, a causal relation between pancreatitis and valproic acid seems to have been established.
Background: Huntington’s Disease (HD) is characterized by choreatic movements. Chorea is treated with antidopaminergic neuroleptics like tiaprid and tetrabenazine. Especially in patients with signs of parkinsonism antidopaminergic medication often leads to a worsening of symptoms. In earlier studies valproic acid showed no beneficial effect on involuntary movements. Myoclonus is rare in HD and ...
We describe a novel isothermal gas-chromatographic procedure for measuring valproic acid. Plasma, with cyclohexanecarboxylic acid added as internal standard, is selectively extracted with pentane to minimize the extraction of other acidic drugs. To convert carboxylic acids to their phenacyl esters, alpha-bromoacetophenone is added to the organic extract before evaporating the solvent. These est...
Background and Aim: Hair loss is one of the most common problems among the skin and hair related diseases. Hair loss can lead to anxiety, depression, low self-esteem and dissatisfaction with appearance. Methods: Patients with androgenetic alopecia who met the eligibility criteria entered the study. Participants were asked to randomly apply a solution of valproic acid 8.3% or a solution of...
objective in this study, we investigated the changes of the serum levels of thyroidhormones including thyroxine (t4), triiodothyronine (t3), t3 resin uptake andthyroid stimulating hormone (tsh) in epileptic children during treatment withanti-epileptic drugs (aeds) including carbamazepine (cbz), primidone (prm),phenobarbital and valproic acid (vpa). materials and methods this study consisted of ...
