چکیده مقاله:

Conventional methods for fungal identification in the clinical laboratory rely on morphological andphysiological tests. These tests often need several days or weeks to complete and are frequentlyunspecific. Molecular identification mostly implies sequencing, which is relatively expensive andtime-consuming, as well as impractical for large numbers of isolates. The Rolling CircleAmplification approach, known as RCA, is a quick, critical and economic method for fungal species'identification. Despite its high speed, this method is highly sensitive, and it has been widely used forthe detection of pathogenic fungi. The specific probes are designed based on the differences in thenucleotide regions of the target gene for the target species. The amplification product can bevisualized by agarose gel electrophoresis, but can also be visualized in gel-free systems usingfluorescence staining of the amplified product by SYBR Green in combination with a UVtransilluminator. Thus, the simplicity, sensitivity, robustness and low costs make RCA an attractivetechnique for the reliable identification of sibling species and other closely related fungi.

use of padlock probes and rolling circle amplification (rca) for rapid identification of trichophyton species, related to human and animal disorder

conclusions species identification of trichophyton is crucially important for epidemiological and phylogenetic purposes and for genotype delineation. rca based on its polymorphisms can be used to generate identification barcodes and as an alternative to dna sequencing; it is a very fast, specific, and economical tool for species identification. results of the 61 putative trichophyton clinical i...

The Discovery of Rolling Circle Amplification and Rolling Circle Transcription.

Nucleic acid amplification is a hugely important technology for biology and medicine. While the polymerase chain reaction (PCR) has been highly useful and effective, its reliance on heating and cooling cycles places some constraints on its utility. For example, the heating step of PCR can destroy biological molecules under investigation and heat/cool cycles are not applicable in living systems....

Improvements of rolling circle amplification (RCA) efficiency and accuracy using Thermus thermophilus SSB mutant protein

Rolling circle amplification (RCA) of plasmid or genomic DNA using random hexamers and bacteriophage phi29 DNA polymerase has become increasingly popular in the amplification of template DNA in DNA sequencing. We have found that the mutant protein of single-stranded DNA binding protein (SSB) from Thermus thermophilus (Tth) HB8 enhances the efficiency of amplification of DNA templates. In additi...

Detection of Rotavirus Using Padlock Probes and Rolling Circle Amplification

Rotavirus infections are one of the most common reasons for hospitalizations due to gastrointestinal diseases. Rotavirus is often diagnosed by latex agglutination assay, chromatography immunoassay, or by electron microscopy, which are all quite insensitive. Reverse transcription polymerase chain reaction, on the other hand, is very sensitive to variations at the genomic level. We developed a no...
