چکیده مقاله:

Abstract Background and Objective: One of the proteins shared in all strains of Brucella is 31 kDa surface protein (BCPS31) that could be an appropriate target for immunization and serological diagnosis. Material and Methods: In the present study, BCSP31 produced as a recombinant protein in pET28a (+) expression system was utilized, using ELISA, to detect trace specific antibody (IgG) in brucellosis patients' serum that was confirmed by culture. We also evaluated cytokine response of peripheral blood lymphocytes to this protein in the cell culture. Results: The results indicated a significant amount of surface protein antibodies (IgG) in the serum of patients with brucellosis. Evaluation of lymphocyte responses to rBCSP31 also showed a significant IL-12 and IFN-γ production in patients’ lymphocyte cultures. Conclusion: These results suggest that BCSP31 can elicit specific humoral and cellular responses during host infection and it can be used in designing immunization and serologic diagnosis systems. Brucella abortus L7/L12 Recombinant Protein Induces Strong Th1 Response in Acute Brucellosis Patients

Background: Because of high morbidity of the brucellosis in humans and the potential use of the microorganism as an agent of biologic warfare, protection of effective vaccines and specific diagnostic reagents become necessary to eradicate brucellosis. Objective: In this study we aimed to investigate the cytokine responses and changes in peripheral blood lymphocyte subgroups of acute brucellosis...

Evaluation of Antibody Response to Polysaccharide Vaccine and Switched Memory B Cells in Pediatric Patients with Inflammatory Bowel Disease

Background/Aims: Inflammatory bowel disease (IBD) is a chronic disease of the gastrointestinal tract, whose etiologies are still unknown. This study was performed to evaluate the humoral immune response in terms of B cell functions in selected IBD patients. Methods: Eighteen pediatric patients with IBD, including 12 cases of ulcerative colitis (UC) and six with Crohn disease (CD), were enrolled...

CFP10: mFcγ2 as a novel tuberculosis vaccine candidate increases immune response in mouse

Objective(s): Despite treatment with antibiotics and vaccination with BCG, tuberculosis (TB) is still considered as one of the most important public health problems in the world. Therefore, designing and producing a more effective vaccine against TB seems urgently. In this study, immunogenicity of a fusion protein which consisting or comprising CFP-10 from Mycobacterium tuberculosis and the Fc-...

Protective efficacy by various doses of a new brucellosis vaccine candidate based on Salmonella strains expressing Brucella abortus BSCP31, Omp3b and superoxide dismutase against brucellosis in murine model

Brucella species are important etiological agents of zoonotic diseases. Attenuated Salmonella strains expressing Brucella abortus BCSP31, Omp3b and superoxide dismutase proteins were tested as vaccine candidates in this study. In order to determine the optimal dose for intraperitoneal (IP) inoculation required to obtain effective protection against brucellosis, mice were immunized with various ...

Assessment of humoral immune response of a Cytomegalovirus DNA-vaccine candidate in BALB/c mice

Introduction: Glycoprotein B (gB) is the major antigen for induction of humoral responses against human cytomegalovirus (HCMV) making it an attractive candidate for immune prophylaxis. In the present study, the humoral immune response of BALB/c mice to a truncated HCMV gB protein fused with GFP was evaluated. Methods: The truncated gB coding sequence was synthesized and cloned in pEGFPN1 eukary...

Construction of an Expression Plasmid (Vector) Encoding Brucella melitensis Outer Membrane Protein, a Candidate for DNA Vaccine

Background: DNA vaccination with plasmid encoding bacterial, viral, and parasitic immunogens has been shown to be an attractive method to induce efficient immune responses. Bacteria of the genus Brucella are facultative intracellular pathogens for which new and efficient vaccines are needed. Methods: To evaluate the use of a DNA immunization strategy for protection against brucellosis, a pla...
