Prostaglandins, Histamine and Platelet Activating Factor: Different Mediators in Dithranol-Induced Skin Damage
نویسندگان
چکیده مقاله:
Dithranol is a potent agent in treating psoriasis but its adverse effects on intact skin have limited its usage. There are many proposed mediators for its adverse effects including prostaglandins, histamine, platelet activating factor and free radicals. In this study we examined the effect of different agents (diazepam, terfenadine, indomethacin and garlic extract) on dithranol-induced skin damage in mice. Animals were treated with different drugs before local application of dithranol cream on the right buttock. Skin biopsy was performed after 48 or 96 hours. Diazepam (3 mg/Kg), terfenadine (20 mg/Kg), indomethacin (25 mg/Kg) and garlic extract (1% and 2%) were all capable of reducing dithranol-induced skin damage. It seems that the role of prostaglandins, histamine and platelet activating factor in the pathogenesis of dithranol-induced skin damage is inevitable. Garlic extract effect might be mediated by platelet activating factor but it needs further work to be proven.

Modulation of Leishmania (L.) amazonensis Growth in Cultured Mouse Macrophages by Prostaglandins and Platelet Activating Factor
The role of endogenously synthesized PAF and prostaglandins on the infection of mouse macrophages by Letsbmanta (L.) amazonensis was investigated, as well as the possible correlation between the effects of these inflammatory mediators with nitric oxide production. It was found that pretreatment of macrophages with 10(-5) M of the PAF antagonists, BN-52021 or WEB-2086, increased macrophage infec...
متن کاملPlatelet Activating Factor and FMLP Induced Intracellular Signaling in Neutrophils
Stimulation of human neutrophils with chemoattractants FMLP or platelet activating factor (PAF) results in different but overlapping functional responses. We questioned whether these differences might reflect patterns of intracellular signal transduction. Stimulation with either PAF or FMLP resulted in equivalent phosphorylation and activation of the mitogen-activated protein kinase (MAPk) homo...

Platelet-activating factor (PAF) is a potent, bioactive phospholipid that acts on multiple cells and tissues through its G protein-coupled receptor (GPCR). PAF is not stored but is rapidly generated via enzymatic acetylation of the precursor 1-O-hexadecyl-2-hydroxy-sn-glycero-3-phosphocholine (lysoPAF). The bioactivity of PAF is effectively and tightly regulated by PAF acetylhydrolases, which c...

This study investigated the mechanisms of platelet-activating factor (PAF)-induced angiogenesis in a mouse model of Matrigel implantation. PAF induced a doseand time-dependent angiogenic response. Inhibitors of nuclear factor (NF) B expression or action, including antisense oligonucleotides to the p65 subunit of NF B (p65 antisense) and antioxidants such as -tocopherol and N-acetyl-L-cysteine, ...

Platelet-activating factor is a phospholipid with diverse potent physiological effects. Its actions are achieved at concentrations as low as IO-‘” M in some systems and almost always by lo-’ M as an intercellular messenger. The molecular structure of PAF was discovered through parallel investigations of two different biological activities: a factor in the blood of rabbits undergoing anaphylaxis...
