چکیده مقاله:

The repair of peripheral nerve injuries is still one of the most challenging tasks and concerns in neurosurgery. Effect of methylprednisolone-laden hydrogel loaded into a chitosan conduit on the functional recovery of peripheral nerve using a rat sciatic nerve regeneration model was assessed. A 10-mm sciatic nerve defect was bridged using a chitosan conduit (CHIT/CGP-Hydrogel) filled with CGP-hydrogel. In authograft group (AUTO) a segment of sciatic nerve was transected and reimplanted reversely. In methylprednisolone treated group (CHIT/MP) the conduit was filled with methylprednisolone-laden CGP-hydrogel. The regenerated fibers were studied within 16 weeks after surgery. The behavioral, functional and electrophysiological studies confirmed faster recovery of the regenerated axons in methylprednisolone treated group compared to CHIT/Hydrogel group (P <0.05). The mean ratios of gastrocnemius muscles weight were measured. There was statistically significant difference between the muscle weight ratios of CHIT/MP and CHIT/Hydrogel groups (P<0.05). Morphometric indices of regenerated fibers showed number and diameter of the myelinated fibers were significantly higher in CHIT/MP than in CHIT/Hydrogel group. Methylprednisolone-laden hydrogel when loaded in a chitosan conduit resulted in improvement of functional recovery and quantitative morphometric indices of sciatic nerve. It may have clinical implications for the surgical management of patients after facial nerve transection injuries.

The Effects of Local Fetal Brain Extract Administration on the Electromyogram of Crushed Sciatic Nerve in Rat

To evaluate the effects of local administration of the fetal brain extract (FBE) on the crushed peripheral nerve, 15 male Wistar rats were used. The right sciatic nerve of 10 rats was locally underwent the tense compression for 30 seconds and then randomly divided into experimental and control (sham) groups. The remained rats were considered as intact animals. Treatment was commenced weekly, by...

Quantification of functional recovery following rat sciatic nerve transection.

Functional recovery following experimental nerve injury has been notoriously difficult to quantify precisely. The current gold standard in the rat sciatic nerve model involves analysis of footprints of the recovering animal, and computation of the sciatic function index (SFI). We performed transection injuries and measured recovery both by walking track analysis and by a newer, simpler, more qu...

Acetyl Salicylic Acid Locally Enhances Functional Recovery after Sciatic Nerve Transection in Rat

Local effect of acetyl salicylic acid (ASA) on peripheral nerve regeneration was studied using a rat sciatic nerve transection model. Forty-five male healthy White Wistar rats were divided into three experimental groups (n = 15), randomly: Sham-operation (SHAM), control (SIL), and ASA-treated (SIL/ASA) groups. In SHAM group after anesthesia left sciatic nerve was exposed through a gluteal muscl...

Transplantation of Olfactory Mucosa Improve Functional Recovery and Axonal Regeneration Following Sciatic Nerve Repair in Rats

Background: Olfactory ensheathing glia (OEG) has been shown to have a neuroprotective effect after being transplanted in rats with spinal cord injury. This study was conducted to determine the possible beneficial results of olfactory mucosa transplantation (OMT) which is a source of OEG on functional recovery and axonal regeneration after transection of the sciatic nerve. Methods: In this study...

P49: The Evaluation of Aqueous Extract of Glycyrrhiza Glabra on Nerve Recovery in the Rat after Sciatic Nerve Injury

Peripheral nerve injury requires a long recovery period, and recovery, once attained, usually is incomplete. Inflammatory procedures may inhibit functional recovery after nerve injury and produce cell death in both the central nervous system and the peripheral nervous system. Since the glycyrrhiza glabra extract has anti-inflammatory effects, it could reduce the severity of injury. The aim of t...

Liquid Metal as Connecting or Functional Recovery Channel for the Transected Sciatic Nerve

In this article, the liquid metal GaInSn alloy (67% Ga, 20.5% In, and 12.5% Sn by volume) is proposed for the first time to repair the peripheral neurotmesis as connecting or functional recovery channel. Such material owns a group of unique merits in many aspects, such as favorable fluidity, super compliance, high electrical conductivity, which are rather beneficial for conducting the excited s...
