P-74: Crocin Repopulate The Leydig and Sertoli Cells in Testis of Cyclophosphamide Treated Adult Mice
نویسندگان
چکیده مقاله:
Background: Increases in the survival rate of men treated with chemotherapeutic drugs and their desire to have children precipitate concerns about the effects of these drugs on germ cells. This study performed to evaluation of protective effects of crocin in oxidative stress induced by cyclophosphamide (CP) on testis. Materials and Methods: Three groups (6 mice in each) of adult mice were used. Control group treated with normal saline via ip, and group 2 treated with CP 15 mg/kg/ week,ip, and group 3 treated with CP along with crocin 200 mg/kg/day,ip. After 35 days samples were took and fixed in 10% formal saline and paraffin sections were prepared and stained by H&E method. Mean distribution of Laydig and Sertoli cells were counted in 1 mm2 field in 5 regions of each slides by latticed objective device. All obtained data were analyzed by SPSS software in ANOVA and Duncan test. Results: Results showed that the mean distribution of Leydig and Sertoli cells in group that treated with CP (9.27 ± 0.34 and 21.86 ± 0.81), significantly was lower than control group (18.99 ± 0.64 and 61.72 ± 3.17), and mean distribution of Leydig and Sertoli cells (14.41 ± 0.37 and 26.19 ± 0.75) significantly were more than CP treated group (p<0.05). Conclusion: This study showed that crocin ameliorate the oxidative stress effects of CP on reproductive organ.

متن کاملEvaluation of antioxidant effects of crocin on sperm quality in cyclophosphamide treated adult mice
Cyclophosphamide (CP) is one of the anti-neoplastic drugs. Despite its numerous clinical applications, it has devastating effects on the testicles and declines the sperm quality in treated patients. This study was aimed to investigate the protective effect of crocin in improving the toxicity induced by CP in reproductive system. In this study, 24 male adult mice (6 to 8 weeks) were randomly div...
متن کاملP-34: Crocin Ameliorate The In Vitro Fertilization (IVF) in Mice Treated with Cyclophosphamide
Background Oxidative stress induced by cyclophosphamide (CP), as a chemotherapeutic agent, causes vulnerability of sperm and declining the fertility capacity. Then in this study the effect of crocin (saffron extract) on amelioration of these side effects was investigated. MaterialsAndMethods In this study 24 mice were divided in three groups (n=8). Control group received normal saline (0.2 ml/d...
متن کاملSertoli Cells Maintain Leydig Cell Number and Peritubular Myoid Cell Activity in the Adult Mouse Testis
The Sertoli cells are critical regulators of testis differentiation and development. In the adult, however, their known function is restricted largely to maintenance of spermatogenesis. To determine whether the Sertoli cells regulate other aspects of adult testis biology we have used a novel transgenic mouse model in which Amh-Cre induces expression of the receptor for Diphtheria toxin (iDTR) s...

متن کاملDHT Germ cells Genital crest Testis Leydig Sertoli
Sex differentiation is a process that starts early during embryogenesis. In males, the SRY gene, located in the short arm of chromosome Y, encodes for the testis-determining factor, which causes the gonad to differentiate into testis. Testicular androgen synthesis by Leydig cells starts from the eighth week of development. It is initially stimulated by placental chorionic gonadotropin (HCG), an...
