P-67: Nano-Selenium Protects Cisplatin-Induced Spermatotoxicty and Chromatin Abnormality Associated with Free Radicals Toxic Stress

چکیده مقاله:
Background: Cisplatin (CIS), an anticancer alkylating agent, induces DNA adducts and effectively cross links the DNA strands and so affects spermatozoa as a male reproductive tract toxicant. Materials and Methods: The present study investigated the cellular/biochemical mechanisms underlying possible protective effect of selenium nano-particles (Nano-Se) as an established strong antioxidant with more bioavailability and less toxicity, on reproductive toxicity of CIS by assessment of sperm characteristics, sperm DNA integrity and chromatin quality. Histopathological analysis of testis and epididymis were performed for assessment of spermatogenic disorders. To determine the role of oxidative stress (OS) in the pathogenesis of CIS gonadotoxicvity, the level of lipid peroxidation (LPO), antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) and peroxynitrite (ONOO) as a marker of nitrosative stress (NS) and testosterone (T) concentration as a biomarkers of testicular function were measured in the blood and testes. Twenty-eight rats were equally divided into four groups. A single IP dose of CIS (7 mg/kg) and protective OP dose of Nano-Se (2 mg/kg/day) were administered alone or in combination. Results: The CIS-exposed rats showed a significant increase in testicular and serum LPO and peroxynitrite level, along with a significant decrease in enzymatic antioxidants levels, diminished serum testosterone concentration and abnormal histologic findings with impaired sperm quality associated with increased DNA damage and decreased chromatin quality. Coadministration of Nano-Se significantly improved the serum testosterone, sperm quality, spermatogenesis and reduced CIS induced free radicals toxic stress and sperm DNA damage. Conclusion: The current study demonstrated that Nano- Se may be useful to prevent CIS-induced gonadotoxicity through its antioxidant potential.

Cisplatin is a widely used chemotherapeutic agent for the treatment of various malignancies. However, its maximum dose is often limited by severe ototoxicity. Cisplatin ototoxicity may require the production of reactive oxygen species (ROS) in the inner ear by activating enzymes specific to the cochlea. Molecular hydrogen was recently established as an antioxidant that selectively reduces ROS, ...

متن کاملGlycosylation protects proteins against free radicals generated from toxic xenobiotics.
Free radicals generated during peroxidase-catalyzed oxidation of two xenobiotics, carcinogenic Sudan I and an anticancer agent ellipticine, easily attack unmodified proteins but not glycoproteins. A significant inverse correlation between the extent of glycosylation of proteins and the degree of binding of Sudan I or ellipticine radicals to these proteins was observed, whereby the protection on...

Background: It is showed that methamphetamine has harmful effects on male reproductive system. The aim of this study was to investigate the effects of methamphetamine administration on testis weight, sperm count, sperm morphology, and chromatin status as well as sperm apoptotis followed by the cytochemical tests in a mouse model. Materials and Methods: Twenty four NMRI male mice were divided in...
متن کاملLipocalin2 Protects Human Embryonic Kidney Cells against Cisplatin–Induced Genotoxicity
Cisplatin is one of the most useful chemotherapeutics which performs its cytotoxic effectvia accumulation of platinum resulting in oxidative stress, and destruction of cell DNA. Thiscould probably cause secondary cancers in healthy tissues. Lipocalin2 (Lcn2) is a protein whichits expression is increased in oxidative stresses. Therefore, the present study was performedto evaluate the protective ...
