چکیده مقاله:

Background Varicocele is the abnormal inflexion and distension of veins of the pampiniform plexus within spermatic cord and is the most common and correctable known risk factor for male infertility. Varicoceles are found in approximately 40% of infertile men, whereas the incidence in the general male population is approximately 15%. Some studies suggest that varicocele is associated with elevated levels of reactive oxygen species (ROS) in sperm and reduced seminal plasma oxidant capacity. It has been postulated that human semen contains a significant amount of glutathione S-transferase (GST)and that the enzyme could attenuate the toxicity of ROS to sperm. The GST gene family produces isoenzyme that is important in protection against oxidative stress and an increase of ROS associated with reduced activity of GST may lead to sperm membrane damage. Our objective was to investigate the genetic polymorphism of the glutathione S-transferase M1 gene (GSTM1) and to assess the oxidative damage in Iranian infertile men with varicoceles. This study was performed on 35 patients with varicocele and 58 control subjects. MaterialsAndMethods Genomic DNA was extracted from peripheral blood of both group. Genotyping was performed via Multiplex PCR and Statistical Analyses were performed using MedCalc software.0h Results The frequency of GSTM1 null (-) genotype was observed to be 22.85% in infertile men with varicocele as against 5.17% in fertile men. Association (OR= 5.43, P=0.026, 95% CI=1.33-22.13) between GSTM1 null (-) genotype and varicocele- induced infertility was confirmed, overall.E Conclusion GSTM1 null (-) genotype may be an important contributor factor in infertile men with varicocele. Larger population-based studies are needed to determine the relation between this deletion and varicocele-associated infertility.

P-213: Mutation Analysis of Mitochondrial ND4L Gene in Iranian Infertile Men with Varicocele

Background: Varicocele is the abnormal tortuosity and dilatation of the veins of the pampiniform plexus within the spermatic cord. Varicocele-related pathology is suspected in infertility as it leads to elevated temperatures in the scrotum and testes, which has a deleterious effect on spermatogenesis. In Varicocele patients, ROS production is enhanced and total antioxidant capacity (TAC) is red...

P-17: Body Mass Index and Sperm Counts Associated with Varicocele in Infertile Men

Background: Varicocele is a progressive disease and etiology of varicocele is not well known. Some studies have been reported that prevalence of varicocele decreases with increasing BMI. The aim of this study was to determine interrelation of varicocoele with height, body mass index (BMI) and sperm counts in infertile men. Materials and Methods: We retrospectively evaluated the data of all pati...

Mitochondrial Genetic Variation in Iranian Infertile Men with Varicocele

Objective Several recent studies have shown that mitochondrial DNA mutations lead to major disabilities and premature death in carriers. More than 150 mutations in human mitochondrial DNA (mtDNA) genes have been associated with a wide spectrum of disorders. Varicocele, one of the causes of infertility in men wherein abnormal inflexion and distension of veins of the pampiniform plexus is observe...

Mutation analysis of TNP1 gene in infertile men with varicocele

BACKGROUND Varicocele is associated with the failure of ipsilateral testicular growth and development, and the symptoms of pain and reduced fertility. The highly condensed structure of the sperm nuclear chromatin is provided by proper expression of Transition Nuclear Protein (TNP) genes, so any dysregulational expression of these genes results in abnormal spermatogenesis and infertility. OBJE...

Spermatozoa protein alterations in infertile men with bilateral varicocele

Among infertile men, a diagnosis of unilateral varicocele is made in 90% of varicocele cases and bilateral in the remaining varicocele cases. However, there are reports of under-diagnosis of bilateral varicocele among infertile men and that its prevalence is greater than 10%. In this prospective study, we aimed to examine the differentially expressed proteins (DEP) extracted from spermatozoa ce...
