P-100: Improved Efficiency of Somatic Cell Nuclear Transfer in Sheep by Persecution of Recipient Oocytes with Brilliant Cresyl Blue Staining
نویسندگان
چکیده مقاله:
Background: The most important factor affecting the efficiency of somatic cell nuclear transfer (SCNT) is the initial quality of the oocyte. Since the abattoir-derived oocytes are heterogeneous in quality, selection of cohort fully grown/competent oocytes is necessary for proper development of SCNT embryos. Materials and Methods: Glucose-6-phosphate dehydrogenase (G6PDH) is actively expressed in growing oocytes, but decreased in activity as oocytes developing to fully grown state. We selected growing and fully grown sheep oocytes based on G6PDH reaction with brilliant cresyl blue (BCB); growing oocytes (unfinished growth phase with abundant G6PDH) degrade BCB and remain a colorless cytoplasm (BCB-) while fully grown oocytes (with reduced G6PDH activity) are unable to degrade BCB leaving a blue cytoplasm (BCB+) after incubation with 26 μM BCB for 45 minutes. BCB- and BCB+ oocytes were separately matured in vitro and their reprogramming capacity and developmental competence were compared after SCNT by using a handmade method. Results: The percentage of cleavage was significantly higher in cloned embryos reconstituted with BCB+ vs. BCB- oocytes (99.33 ± 0.66% vs. 86 ± 1.15%, respectively). Notably, cloned blastocyst development rate was significantly higher in BCB+ vs. BCB- oocytes (15.66 ± 1.2% vs. 6.33 ± 0.88%, respectively). Conclusion: BCB staining test of immature oocytes is a useful method for selecting competent oocytes and improve SCNT in sheep through increasing the reprogramming ability of the cytoplasm.

Sheep oocytes derived from the ovaries collected from the slaughterhouse are often used for research on in vitro embryo production, animal cloning, transgenesis, embryonic stem cells, and other embryo biotechnology aspects. Improving the in vitro culture efficiency of oocytes can provide more materials for similar studies. Generally, determination of oocyte quality is mostly based on the layers...
متن کاملEfficiency of Ovine Fibroblast or Cumulus Cells For Somatic Cell Nuclear Transfer in Sheep
Purpose: Despite remarkable progresses have been achieved in the field of somatic cell nuclear transfer (SCNT), there is little information regarding the effect of donor cell type on the efficiency mammalian somatic cell cloning in vitro. This study compared in vitro developmental competency of sheep enucleated oocytes reconstructed with either fibroblast or cumulus cells. Material and methods...
متن کاملBrilliant Cresyl Blue in Sap of Nitella
It has been shown by several investigators I that when the pl-I value of the external dye solutions is increased, the rate of accumulation of a basic dye in the cell sap is accelerated. Two different explanations for this have been given. Some have accounted for it on the basis that the basic dye enters the cell in the form of a dye hydrate and combines with the substances in the cell; and that...
متن کاملP-81: Distinct Epigenetic Status of Fully Grown and Growing Oocytes Selected by BCB Staining
Background: Since the sheep ‘Dolly’ was successfully cloned by somatic cell nuclear transfer (SCNT), the inefficiency of SCNT cloning and abnormality of cloned animals have been a focus of considerable controversy. Oocyte quality has been shown to contribute to poor somatic cloning and cell reprogramming efficiency with many factors affecting oocyte quality and developmental potential. It has b...
متن کاملDevelopmental competence of prepubertal goat oocytes selected with brilliant cresyl blue and matured with cysteamine supplementation.
The aim of this study was to assess the effect of oocyte selection using the brilliant cresyl blue (BCB) test plus the addition of cysteamine to the in vitro maturation (IVM) medium to improve the in vitro embryo development of prepubertal goat oocytes. The oocytes were exposed to 26 microM BCB and classified according to their cytoplasm coloration: BCB+ (oocytes with blue cytoplasm) and BCB- (...
متن کاملI-12: Nuclear Reprogramming in Bovin Somatic Cell Nuclear Transfer
Somatic cell nuclear transfer (SCNT or cloning) returns a differentiated cell to a totipotent status; a process termed nuclear reprogramming. Reproductive cloning has potential applications in both agriculture and biomedicine, but is limited by low efficiency. To understand the deficiencies of nuclear reprogramming, our research has focused on both candidate genes and global gene expression pat...
