O-19: Fertilization Rates Are Improved after IVF if The Corona Radiata Is Left Intact in Vitrified-Warmed Human Oocytes
نویسندگان
چکیده مقاله:
Background: Before human MII oocytes are vitrified they are usually denuded from their cumulus cells. In this study we wanted to investigate the effects of an intact corona radiata on the vitrification and fertilization of human oocytes. Materials and Methods: The study comprised two different parts. In Part 1, 36 MII stage oocytes, from 6 patients, were randomly assigned into a control group, a group of vitrified-warmed oocytes without a corona radiata and a group of vitrified-warmed oocytes with an intact corona radiata. In each group of 12, 6 oocytes were used for evaluation of the zona pellucida solubility (hardening) and another 6 oocytes were used for the analysis of their ultrastructure. In addition, six polyspermically fertilized oocytes were used as positive controls for zona pellucida hardening. In Part 2, 16 patients in total produced 107 fresh and 98 vitrified-warmed oocytes, with or without an intact corona radiata. All oocytes were fertilized via conventional IVF and embryos were transferred according to our standard ET routines. The oocyte survival and fertilization rates, embryo quality and pregnancy and implantation rates were evaluated. Results: There were no differences in oocyte survival, zona pellucida solubility (hardening) or the number of cortical granules between the vitrified-warmed and fresh oocytes. There were also no differences in the zona pellucida solubility and the number of cortical granules between vitrified-warmed oocytes with or without an intact corona radiata. However, the oocytes with an intact corona radiata had a higher fertilization rate after conventional IVF insemination. No differences were seen in the survival and cleavage rates, the percentage of highquality embryos or the clinical outcome. Conclusion: Zona hardening and ultrastructural damage do not seem to occur in vitrified human oocytes. An intact corona radiata in vitrified-warmed oocytes retains their fertilization capacity in conventional IVF, but does not improve the embryo quality. Poor fertilizing capacities of vitrified-warmed oocytes without an intact corona radiata seem to have been due to the complete removal of the cumulus cells.

متن کاملRapamycin Influences the Efficiency of In vitro Fertilization and Development in the Mouse: A Role for Autophagic Activation
The mammalian target of rapamycin (mTOR) regulates cellular processes such as cell growth, metabolism, transcription, translation, and autophagy. Rapamycin is a selective inhibitor of mTOR, and induces autophagy in various systems. Autophagy contributes to clearance and recycling of macromolecules and organelles in response to stress. We previously reported that vitrified-warmed mouse oocytes s...
متن کاملO-41: Comparison of Vitrification Systems inHuman Oocytes and Evaluation of HSP A1Aand MnSOD Gene Expression Patterns
Background: The aim of this study was to compare two vitrification systems (Cryotop and OPS) on mouse MII oocytes and evaluate its effects on expression of heat shock protein A1A and MnSOD genes. Materials and Methods: Groups of about 15 oocytes without cumulus complexes (granulose, et. al.) were collected and exposed to equilibration and vitrification solutions before loading the oocytes into ...
متن کاملStudy of the Zona Radiata Structure in Oocytes of the Persian Sturgeon (Acipenser persicus) before and after Fertilization
Egg envelope or otherwise called zona radiata (ZR) is an acellular area that originates next to oolemma externally. It grows and thickens gradually from oolemma and follicular epithelium both. Microscopic studies on the tissue sections hava shown that during developmental stages of oocytes, ZR could not be observed at stage I (Primary growth stage) in Acipenser persicus. It appeared in stage ...
متن کاملLaser-Assisted In Vitro Fertilization Facilitates Fertilization of Vitrified-Warmed C57BL/6 Mouse Oocytes with Fresh and Frozen-Thawed Spermatozoa, Producing Live Pups
The utility of cryopreserved mouse gametes for reproduction of transgenic mice depends on development of assisted reproductive technologies, including vitrification of unfertilized mouse oocytes. Due to hardening of the zona pellucida, spermatozoa are often unable to penetrate vitrified-warmed (V-W) oocytes. Laser-assisted in vitro fertilization (LAIVF) facilitates fertilization by allowing eas...
متن کاملClinical pregnancy from a vitrified/warmed human blastocyst.
The first pregnancy after vitrification of a human blastocyst (day 5 of embryo culture) was reported by Yokota et al. in 2000. Since then more attention has been given to the technique of vitrification and its safe application in ART. To the best of our knowledge, this is the first report of a clinical pregnancy resulting in a live birth from the transfer of a vitrified/ warmed human blastocyst...
