چکیده مقاله:

Background: Recent studies have revealed that adiponectin, a novel adipocytokine, and its two receptors (AdipoR1 and AdipoR2) are involved in regulation of ovarian function in various farm animal species. The aim of this study was to investigate the gene expression pattern of Adiponectin and its receptors in ovarian cells of dominant (DFs) and non- dominant follicles (NDFs) in the bovine. Materials and Methods: Based on the estradiol/progesterone ratio, two largest follicles from ovaries collected at a slaughterhouse were classified as NDFs and DFs. In addition, the stage of the estrous cycle (follicular or luteal phases) of the slaughtered cows was determined by macroscopic observation of the ovaries and the uterus. The relative expression of adiponectin, AdipoR1 and AdipoR2 mRNA in theca and cumulus cells were determined by qRT-PCR. Results: Adiponectin and its receptors genes were clearly expressed higher (p<0.05) in theca and cumulus cells of DFs than those of NDFs during the follicular and luteal phases of bovine estrous cycle. Positive correlation (r=0.7; p<0.01) was observed between adiponectin mRNA level in ovarian cells of DFs and follicular fluid estradiol concentration in follicular phase. Adiponectin mRNA abundance in ovarian cells of NDFs showed a significant negative correlation with follicular fluid progesterone concentration in follicular phase (r= -0.7; p<0.01) and luteal phase (r= -0.80, p=0.01). Conclusion: This work has revealed the novel association of adiponectin and its receptors genes with follicular dominance.

Association of Adiponectin rs1501299 Gene Polymorphism with Adiponectin Levels and Type 2 Diabetes in an Iranian Population

Background and Objectives: Adiponectin is an adipokine, which is abundantly expressed in adipose tissue and has a potent roles in insulin sensitivity. This study aimed to test the association of single nucleotide polymorphism of rs1501299 of the adiponectin gene with adiponectin levels and type 2 diabetes.   Methods: This case-control study was conducted on 80 diabetic patients with fasting bl...

Expression of adiponectin receptors and effects of adiponectin isoforms in mouse preimplantation embryos.

BACKGROUND Adiponectin, a pleiotropic hormone secreted from adipose tissue, can mediate some negative effects of obesity on female health, and can participate in the impaired reproductive performance of obese women. Using a mouse model, we investigated expression of adiponectin receptors in ovulated oocytes and in vivo derived preimplantation embryos, and tested effects of different adiponectin...

Insulin regulates the expression of adiponectin and adiponectin receptors in porcine adipocytes.

Adiponectin is an adipocyte-derived hormone that can improve insulin sensitivity. Its functions in regulating glucose utilization and fatty acid metabolism in mammals are mediated by two subtypes of adiponectin receptors (AdipoR1 and AdipoR2). This study was conducted to determine the effect of insulin on the expression of adiponectin and its receptors. We demonstrated that in the presence of 1...

Adiponectin, adiponectin receptors, and epigenetic regulation of adipogenesis.

We studied the molecular mechanism of obesity-induced insulin resistance and adipogenesis. Plasma adiponectin and adiponectin receptor (AdipoR1) in muscle are down-regulated in obesity. Analysis of muscle-specific AdipoR1 knockout mice revealed the pivotal role of adiponectin/AdipoR1 in the regulation of mitochondrial biogenesis via AMPK- and SIRT1-mediated PGC-1α activation as well as Ca(2+)-d...
