Klotho Protein,A Biomarker for AKI
نویسندگان
چکیده مقاله:
Klotho is an anti-aging single-pass membrane protein that is mainly produced in the kidney. The level of soluble klotho decreases with age and the klotho gene is associated with an increased risk of age-related diseases, such as diabetes, skin atrophy, chronic kidney disease, ataxia and cancer. The klotho gene is composed of five exons and encodes a membrane glycoprotein located in the plasma membrane. The 3 forms of klotho protein have a specific function: 1-klotho forms a complex membrane protein with fibroblast growth factor receptors (FGFR) and acts as a binding cofactor for FGF23, which is involved in the regulation of aging and the development of chronic diseases through the regulation of ionized phosphorus and vitamin D metabolism. 2- Secretory klotho as a humoral agent has pleiotropic activity which includes regulation of oxidative stress, growth factor signaling and ion homeostasis. Secretory klotho also plays a role in organ protection. 3- The intracellular form of Klotho suppresses inflammation caused by cellular aging and mineral metabolism.

Acute Kidney Injury (AKI) biomarker.
The kidney has a remarkable capacity to withstand insults for an extended period of time. The sensitivities of individual renal cells to injury vary depending on their type, position in the nephron, local vascularization, and the nature of injury. The resulting kidney injury is a product of the interplay between cell dysfunction, cell death, proliferation, inflammation, and recovery. The Acute...

262 Am J Clin Pathol 2013;139:259-262 © American Society for Clinical Pathology for us is to try to compare sKL levels in normal people and patients with cancer. However, the concern with this is that because the sKL level could be affected by many factors such as age, diabetes, kidney disease, and others,5 in patients with cancer the sKL level would vary because of such factors, in addition to...
متن کاملHydrogen-Rich Saline Alleviates Kidney Fibrosis Following AKI and Retains Klotho Expression
Purpose: Acute kidney injury (AKI) is a prominent risk factor for the development of chronic kidney disease (CKD). To date, the related mechanism and effective therapy have not been rigorously explored. The present study aims to investigate the reno-protection of hydrogen-rich saline (HRS) against ischemia/reperfusion (IR)-induced AKI. Methods: Adult male C57 mice were randomly allocated into t...
متن کاملKlotho: a novel and early biomarker of acute kidney injury after cardiac valve replacement surgery in adults.
Klotho is a potential biomarker and therapeutic target in a model of acute kidney injury (AKI) induced in rats by ischemia-reperfusion injury. However, the sensitivity and specificity of serum Klotho for early detecting clinical AKI are unknown. This prospective study evaluated the significance of serum Klotho for early detection of postoperative AKI among adult patients undergoing cardiac valv...

متن کاملSubclinical AKI is still AKI
The concept of acute kidney syndromes has shifted in recent years from acute renal failure to acute kidney injury (AKI). AKI implies injury or damage but not necessarily dysfunction. The human kidney has an important glomerular function reserve, and dysfunction becomes evident only when more than 50% of the renal mass is compromised. Recent AKI classifications include even slight changes in ser...
