Human chorionic gonadotropin attenuates amyloid-β plaques induced by streptozotocin in the rat brain by affecting cytochrome c-ir neuron density

چکیده مقاله:
Objective(s): Amyloid β plaques, in Alzheimer’s disease, are deposits in different areas of the brain such as prefrontal cortex, molecular layer of the cerebellum, and the hippocampal formation. Amyloid β aggregates lead to the release of cytochrome c and finally neuronal cell death in brain tissue. hCG has critical roles in brain development, neuron differentiation, and function. Therefore, we investigated the effect of hCG on the density of the congophilic Aβ plaque and cytochrome c-ir neurons in the hippocampus, prefrontal cortex, and cerebellum of Streptozotocin (STZ)-treated rats. Materials and Methods: Alzheimer model in rats (except the control group) was induced by streptozotocin (3 mg/kg, Intracerebroventricularly (ICV)). Experimental group rats received streptozotocin and then different doses of hCG (50, 100, and 200 IU, intraperitoneally) for 3 days. 48 hr after last drug injection and after histological processing, the brain sections were stained by congo red for congophilic amyloid β plaques and cytochrome c in the hippocampus, prefrontal cortex, and cerebellum were immunohistochemically stained. Results: Density of congophilic Aβ plaques and cytochrome c-immunoreactive neurons was significantly higher in ICV STZ treated rats than controls. Treatment with three doses of hCG significantly decreased the density of congophilic Aβ plaques and cytochrome c-immunoreactive neurons in the rat hippocampus, prefrontal cortex, and cerebellum in ICV STZ-treated rats (P

Human chorionic gonadotrophin (hCG) is a hormone produced during pregnancy and present at the implantation site and in the maternal blood. Pregnancy has been proposed to represent a controlled state of inflammation at an early stage at the implantation site and later, systemically extended to the maternal circulation. Earlier, we reported that hCG can inhibit the development of diabetes in NOD ...
متن کاملEffect of Epigallocatechin Gallate and Catechin on Overexpression of GSK-3β and IR Genes Induced by Streptozotocin in Rat Brain
Background: Type 2 diabetes mellitus (T2DM) is one of the significant risk factors for Alzheimer disease (AD). Defects in insulin signaling pathway induce AD hallmarks mainly through activation of glycogen synthase kinase-3β (GSK-3β) pathway. Objectives: In this study, we investigated the expression of GSK-3β and insulin receptor (IR) genes in the hippocampi of an animal model of sporadic AD ...
متن کاملQuantification of Human Chorionic Gonadotropin by Bovine Serum Albumin Nanoparticles
Background and Aims: Some nanoparticles can be used in immunoassays to increase sensitivity. This study aimed to evaluate a novel nano-immunoassay based on bovine serum albumin nanoparticles (BSA NPs). Materials and methods: At first, the nanostructure was synthesized, and then applied as a tag in the nano-immunoassay. Then the concentration of β-subunit of human chorionic gonadotropin ...
متن کاملDiosgenin attenuates cardiac oxidative stress in streptozotocin- induced diabetic rat
Background and Objective: Chronic diabetes mellitus (DM) leads to cardiovascular dysfunction. Diosgenin is a natural steroidal saponin with cardiovascular protective potential. In this research study, the beneficial effect of diosgenin was evaluated on some markers of oxidative stress in cardia tissue of streptozotocin (STZ)-diabetic rats. Materials and Methods: Male Wistar rats (n = 28) were ...
متن کاملEFFECT OF β-HUMAN CHORIONIC GONADOTROPIN (β-HCG) ON NEUTROPHIL FUNCTIONS
Many findings point to the presence of a close relationship between the immune, endocrine and neurologic systems. For example the suppressive effects of β-human chorionic gonadotropin (β-HCG) on IL-l production, the potentiative effect of enkephalins on lymphocyte functions and the suppressive effect of leuteinizing hormone (LH) on natural killer (NK) cell activities have been clearly esta...
