چکیده مقاله:
Introduction: [18F]-6-thia-14-fluoro-heptadecanoic acid 3b, a free fatty acid, has been used in myocardial PET imaging. In order to establish an automated synthesis module for routine production in the country, a study performed for optimization of the production conditions as well as making modifications. Methods: [18F] Benzyl-14-Fluoro-6-thia-heptadecanoate 2b was prepared in no-carrier-added (n.c.a) form from Benzyl-14-tosyloxy-6-thia-heptadecanoate 1 in one step at 90C in Kryptofix2.2.2/[18F] and acetonitrile as the solvent followed by Silica column chromatography. The radiolabeled ester 2 was then hydrolyzed to yield [18F]-6-thia-14-fluoro-heptadecanoic 3b. The final solution was concentrated using C18 SPE system and administered to normal rats for biodistribution as well as co-incidence imaging studies. Results: The synthesis took 15 min with overall radiochemical yield of 15-25% (EOS) and chemical-radiochemical purity more than 94%. Automation was performed using a two-pot synthesis. The best imaging time was shown to be 140-180 minutes post injection. Conclusions: Using this procedure a fast, reliable, automated synthesis for the cordial PET tracer, i.e. [18F]-FTHA can be obtained without HPLC purification step.

متن کاملFatty acid uptake is preserved in chronically dysfunctional but viable myocardium.
Glucose uptake appears preserved or even enhanced in the chronically dysfunctional but viable myocardium. However, the use of other fuels such as free fatty acids (FFA) remains unknown. We studied FFA uptake in the chronically dysfunctional but viable myocardium in seven patients with an occluded major coronary artery and a corresponding chronic wall motion abnormality but no previous infarctio...
متن کاملLongitudinal Evaluation of Myocardial Fatty Acid and Glucose Metabolism in Fasted and Nonfasted Spontaneously Hypertensive Rats Using MicroPET/CT
Using longitudinal micro positron emission tomography (microPET)/computed tomography (CT) studies, we quantified changes in myocardial metabolism and perfusion in spontaneously hypertensive rats (SHRs), a model of left ventricular hypertrophy (LVH). Fatty acid and glucose metabolism were quantified in the hearts of SHRs and Wistar-Kyoto (WKY) normotensive rats using long-chain fatty acid analog...
متن کاملProduction and quality control of [18F]-FDG in Iran [Persian]
The use of [18F]-2-FDG has been increasing in recent years. This agent was originally used for evaluation of CNS glucose uptake and pathologic conditions by positron emission tomography (PET). In view of increasing demand for this agent, its production has been started at cyclotron dept., Nuclear research center for agriculture & Medicine-Karaj Iran (NRCAM). In this paper, [18F]-FDG synth...
متن کاملA within-subject comparison of 6-[18F]fluoro-m-tyrosine and 6-[18F]fluoro-L-dopa in Parkinson's disease.
Progression of Parkinson's disease symptoms is imperfectly correlated with positron emission tomography biomarkers for dopamine biosynthetic pathways. The radiopharmaceutical 6-[(18) F]fluoro-m-tyrosine is not a substrate for catechol-O-methyltransferase and therefore has a more favorable uptake-to-background ratio than 6-[(18) F]fluoro-L-dopa. The objective of this study was to evaluate 6-[(18...
متن کاملEvaluation of trans-9-18F-fluoro-3,4-Methyleneheptadecanoic acid as a PET tracer for myocardial fatty acid imaging.
UNLABELLED This study describes the radiosynthesis and preliminary biologic evaluation of trans-9(RS)-(18)F-fluoro-3,4(RS,RS)-methyleneheptadecanoic acid ((18)F-FCPHA) as a new potential probe for assessing myocardial fatty acid metabolism by PET. This fatty acid, containing a cyclopropyl moiety in the beta,gamma-position, was designed to enter the myocardium by the same mechanism as natural fa...
