چکیده مقاله:
Background: The cuprizone multiple sclerosis (MS) animal model is characteristic for toxic demyelination and represents a reversible demyelination and remyelination system. It has been shown that green tea epigallocatechin-3-gallate (EGCG) might be effective in improving the symptoms and pathological conditions associated with autoimmune inflammatory diseases in several animal models. Objectives: In this study the effects of EGCG on proteolipid protein (PLP) and oligodendrocyte transcription factor 1 (Olig1) expression in the cerebral cortex of murine model of cuprizone-induced demyelination was investigated. Materials and Methods: C57BL/6 mice were treated by cuprizone for six weeks in order to induce demyelination. Immediately after the cessation of cuprizone the animals were divided into 6 groups (n=10 for each groups). The first two groups was injected intraperitoneally (IP) by EGCG in the amount of 50 mg/kg /daily body weight for 2 and 4 weeks. The second two groups (SHAM) was injected IP by phosphate-buffered saline (PBS) for 2 and 4 weeks and the third two groups was left without injection as controls. After two and four weeks the mice were killed and the cerebral cortex was collected and the expression of PLP and Olig1 was studied by Western blotting. Results:The results showed that there is a significant increases in PLP and Olig1 expression among the EGCG treated group as compared to the SHAM and control groups (p<0.0001). Conclusion: It is concluded that EGCG increases PLP and Olig1 expression in the cerebral cortex of mouse model of MS induced by cuprizone.

متن کاملThe enhancing effect of electromagnetic field on the expression of Oligodendrocyte transcription factor 1 and 2 (Olig1/2) in the mice cerebral cortex
Olig1 and Olig2, two transcription factors, play regulatory function in the differentiation and specification of oligodendrocyte progenitor cells (OPCs). In this study the effects of electromagnetic fields (EMF) on total protein concentration ( TPC ) and Olig1 and Olig2 expression in the cerebral cortex of mouse was examined. Twenty-one Balb/c mice were separated into three groups: control, EMF...

Background and purpose: Oligodendrocytes are responsible for myelin synthesis in the central nervous system (CNS). Olig1 and Olig2 play an important role in regulating the development of oligodendrocyte precursor cells (OLPs). Myelin basic protein (MBP) is the main component of myelin sheath. Leukemia inhibitory factor (LIF) has an important role in myelination and pathology of multiple scleros...

Alzheimer's disease (AD) is a progressive neurodegenerative disorder pathologically characterized by deposition of beta-amyloid (Abeta) peptides as senile plaques in the brain. Recent studies suggest that green tea flavonoids may be used for the prevention and treatment of a variety of neurodegenerative diseases. Here, we report that (-)-epigallocatechin-3-gallate (EGCG), the main polyphenolic ...

Naturally occurring polyphenols are potent antioxidants. Some of these compounds are also ligands for the GABA(A) receptor benzodiazepine site. This feature endows them with sedative properties. Here, the anxiolytic activity of the green tea polyphenol (-)-epigallocatechin gallate (EGCG) was investigated after acute administration in mice, using behavioral tests (elevated plus-maze and passive ...

The most abundant polyphenol in green tea, epigallocatechin-3-gallate (EGCg), has recently received considerable attention due to the discovery of numerous health-promoting bioactivities. Despite reports of its poor oral bioavailability, EGCg has been included in many dietary supplement formulations. Conventional preformulation methods have been employed to improve the bioavailability of EGCg. ...
