Down regulation of GTPase regulator associated with the focal adhesion kinase (GRAF) gene expression in patients with acute myeloblastic leukemia
نویسندگان
چکیده مقاله:
Introduction: GTPase regulator associated with focal adhesion kinase (GRAF) is a recently identified GTPase activating protein that has the tumor suppressor properties. However, the expression level of GRAF in leukemia had received less attention. The main purpose of this research was the evaluating of the expression level of GRAF in patients with acute myeloid leukemia (AML). Materials and methods: Ninety people participated in the research (45 patients with AML and 45 healthy persons). RNA was extracted from their blood samples and cDNA synthesized. The expression levels of GRAF mRNA were determined by QRT-PCR method. Results: The expression levels of GRAF mRNA were significantly lower in AML samples than the control groups. WBC count was high in down-regulated samples than up-regulated ones (P< 0.05). There was any significant difference between two groups in other laboratories parameters. Conclusion: In spite of rare studies on GRAF gene expression level in patients with AML, was shown that it's down-regulated in people with AML.

متن کاملGTPase regulator associated with the focal adhesion kinase (GRAF) transcript was down-regulated in patients with myeloid malignancies
BACKGROUND GTPase regulator associated with the focal adhesion kinase (GRAF), a putative tumor suppressor gene, is found inactivated in hematopoietic malignancies by either genetic or epigenetic abnormalities. However, the expression level of GRAF gene has not yet been studied in leukemia. The aim of this study was to investigate the expression level of GRAF gene in those patients with myeloid ...
متن کاملارزیابی میزان بیان ژن GRAF در بیماران B-ALL مراجعه کننده به بیمارستانهای مشهد و ارتباط آن با یافتههای آزمایشگاهی و بالینی بیماران
Background and Objective: Acute lymphoblastic leukemia is a disorder caused by the proliferation of malignant immature lymphoid cells. GRAF (GTPase regulator associated with focal adhesion kinase) which acts as a tumor suppressor gene is disabled in hematologic malignancies due to genetic and epigenetic abnormalities. In this study, GRAF gene expression was inspected in patients with B-ALL. ...
متن کاملOver-expression of Wilm’s Tumor Gene 1 (WT1) in Iranian Patients with Acute Myeloblastic Leukemia
Background: The Wilm’s tumor gene 1 (WT1) encodes a zinc finger transcription factor that is inactivated in a subset of Wilm’s tumors. It plays a crucial role in growth, proliferation and development of some embryonic and adult organs. WT1 is expressed as a tumor associated antigen (TAA) in various types of solid and hematopoietic malignancies and can be employed as a useful marker for targeted...
متن کاملExpression of ROR1 Gene in Patients with Acute Lymphoblastic Leukemia
Background: Acute lymphoblastic leukemia (ALL) results from genetic alterations in a single lymphoid progenitor cell. Expression of ROR1 is reported to be increased in ALL and mantle cell lymphoma. In this study the expression of ROR1 was assessed in newly diagnosed patients with ALL. Methods: This study was carried out on 40 patients with newly diagnosed ALL and healthy individuals as contro...
متن کاملCytoskeletal changes induced by GRAF, the GTPase regulator associated with focal adhesion kinase, are mediated by Rho.
Graf, the GTPase regulator associated with focal adhesion kinase was previously shown to have GAP activity for &Rgr; A and Cdc42 in vitro (Hildebrand et al 1996 Mol. Cell Biol. 16: 3169-3178). In this study we sought to determine whether Graf acted at the level of Cdc42, Rho, or both in vivo and whether Graf was a signal terminator or transducer for these proteins. Microinjection of Graf cDNA i...
