Conditioned Medium from Cultured Colorectal Cancer Cells Affects Peripheral Blood Mononuclear Cells Inflammatory Phenotype in Vitro
نویسندگان
چکیده مقاله:
Background: Colorectal cancer (CRC) is the third most common cancer worldwide. Studies have indicated that immune cells and soluble factors play a key role in maintaining the balance between tumor-promoting inflammation and anti-tumor immunity. It has been shown that secreted cytokines from CRC cell lines could affect peripheral blood mononuclear cells (PBMCs), monocytes, and macrophages phenotypes. Macrophage infiltration has been associated with good prognosis in some cancers, but with poor prognosis in others. The present study aimed to evaluate the effect of conditioned media from CRC cells (Caco-2) on immune responses produced by PBMCs. Methods: The present study was performed at the Gastroenterology and Liver Diseases Research Institute, Shahid Beheshti University of Medical Sciences (Tehran, Iran) in 2017. Human monocytes were isolated from PBMCs by Ficoll gradient media. The co-culture of monocytes and Caco-2 conditioned media was carried out. RNA extraction and cDNA synthesis of monocytes were performed after 96 hours. Gene expression of pro- and anti-inflammatory cytokines was evaluated by real-time PCR. Statistical analysis was performed using the SPSS software (version 21.0) with the independent sample t test. P

Response of peripheral blood mononuclear cells to conditioned medium from cultured oral squamous cell carcinomas.
The current study investigated the capacity for tumor factors secreted by head and neck squamous cell carcinoma (HNSCC) cell lines, KB, KB16, and HEP, to induce the secretion of various cytokines from peripheral blood mononuclear cells (PBMCs). PBMCs were isolated from blood samples collected from six healthy volunteers and these cells were incubated for 6, 24, 48, or 72 hours in the presence o...
متن کاملEvaluation of in vitro Effect of Pyocyanine Pigment on Interleukin-2 Secretion from Peripheral Blood Mononuclear Cells in Cancer Patients
Background and Objective: The pyocyanine pigment in Pseudomonas aeruginosa stimulates blood cells to secrete IL-2. IL-2 cytokine is an activator of cytotoxic T cells and natural killer cells. These cells destroy the target cells of patients with cancer. This study aimed to evaluate the effects of pyocyanine on the IL-2 secretion from peripheral blood mononuclear cells (PBMCs) in patients with b...

Background and Aim: Today, cancer is considered as a major health problem and affects the health of society. Breast cancer is the second leading cause of cancer death in women after lung cancer. According to epidemiological studies, cancer is the second most common cause of death after cardiovascular disease worldwide and the third leading cause of death after cardiovascular disease and acciden...
متن کاملOptimization of human peripheral blood mononuclear cells (PBMCs) cryopreservation
Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...
متن کاملPeripheral Blood Mononuclear Cells
Numerous cell types are involved in maintenance of the intestinal tissue. However, the main players are cells of the epithelial lining and the immune system. Human peripheral blood mononuclear cells (PBMCs) are used to investigate the effect of food bioactives on various immune cells. These cells are easily isolated from blood of healthy donors or buffy coats (leukocyte concentrates, a by-produ...
متن کاملIrradiated cultured apoptotic peripheral blood mononuclear cells regenerate infarcted myocardium.
BACKGROUND Acute myocardial infarction (AMI) is followed by post AMI cardiac remodelling, often leading to congestive heart failure. Homing of c-kit+ endothelial progenitor cells (EPC) has been thought to be the optimal source for regenerating infarcted myocardium. METHODS Immune function of viable peripheral blood mononuclear cells (PBMC) was evaluated after co-culture with irradiated apopto...
