Association between Insulin–Like Growth Factor I Polymorphism and Early Growth Traits in Iranian Zandi Sheep, Found Polymerase Chain Reaction Restriction Fragment Length Polymorphism (PCR-RFLP)

چکیده مقاله:
Early growth traits in sheep are economically important. Breeding for these traits has been shown to be problematic using quantitative genetic methods, particularly in native sheep herds. Molecular genetics is useful for sheep breeding. The gene for insulin-like growth factor 1 (IGF1) is one of the important candidate genes known for ovine early growth traits. The aim of this study was to investigate IGF1 polymorphism in exon 1 and its association with some early growth traits in Iranian Zandi sheep. For this purpose, 120 male and female lambs were selected randomly at up to 5 months old and blood samples were taken individually. Also, phenotypic records were collected from related farms. Genomic DNA was extracted by a salting-out method. A 265 bp region in exon 1 of IGF1 was amplified by polymerase chain reaction (PCR), then genotyped using the restriction endonuclease HaeII. Two alleles, here called A and B, were found. Examination using PopGene32 showed allele frequencies of 0.47 A and 0.53 B. Genotype % frequencies were 28.33 AA, 37.50 AB and 34.17 BB. Chi-squared and G-squared tests showed that the population was not at Hardy-Weinberg equilibrium (P

Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) for rapid diagnosis of neonatal sepsis
BACKGROUND & OBJECTIVES The difficulties in diagnosis of neonatal sepsis are due to varied clinical presentation, low sensitivity of blood culture which is considered the gold standard and empirical antibiotic usage affecting the outcome of results. Though polymerase chain reaction (PCR) based detection of bacterial 16S rRNA gene has been reported earlier, this does not provide identification o...
متن کاملRestriction Fragment Length Polymorphism (RFLP)
Restriction Fragment Length Polymorphism (RFLP) is a technique in which organisms may be differentiated by analysis of patterns derived from cleavage of their DNA. If two organisms differ in the distance between sites of cleavage of a particular restriction endonuclease, the length of the fragments produced will differ when the DNA is digested with a restriction enzyme. The similarity of the pa...
متن کاملDetection of avian reoviruses causing tenosynovitis in breeder flocks in Iran by reverse transcription-polymerase chain reaction (RT-PCR) and restriction enzyme fragment length polymorphism (RFLP)
BACKGROUND:Avian reoviruses (ARVs) are members of theOrthoreovirus genus; one of the 12 genera of the Reoviridaefamily. The ARVs are the cause of some important diseases inpoultry such as reovirus-induced arthritis, tenosynovitis, chronic respiratory disease, and mal-absorption syndrome.OBJECTIVES: In this study, the presence of ARVs in the Iranianbreeder flocks was investigated through reverse ...

متن کاملIntraspecies polymorphism of Cryptosporidium parvum revealed by PCR-restriction fragment length polymorphism (RFLP) and RFLP-single-strand conformational polymorphism analyses.
A glycoprotein (Cpgp40/15)-encoding gene of Cryptosporidium parvum was analyzed to reveal intraspecies polymorphism within C. parvum isolates. Forty-one isolates were collected from different geographical origins (Japan, Italy, and Nepal) and hosts (humans, calves, and a goat). These isolates were characterized by means of DNA sequencing, PCR-restriction fragment length polymorphism (PCR-RFLP),...
متن کاملGenotypic analysis of mutations in Taq I restriction recognition sites by restriction fragment length polymorphism/polymerase chain reaction.
Point mutations in somatic cells play a role in the etiology of several classes of human pathologies. Experimental procedures are required that allow the detection and quantitation of such mutations in disease-related genes in tissue biopsy samples without the need for the selection of mutated cells. We describe the genotypic analysis of single base pair mutations in the Taq I endonuclease reco...
