چکیده مقاله:

ABSTRACT        Background and Objective: Measurement of amino acids is an important tool for metabolic studies and evaluation of patients’ clinical condition. The aim of this study was to analyze the plasma amino acids using reverse phase high performance liquid chromatography techniques (RP-HPLC) with pre-column derivatization by o-phthalaldehyde (OPA) in combination with 3- mercaptopropionic acid (3-MPA).        Methods: Overall, 107 neonates and babies suspected of having metabolic disorder were enrolled in this study. The level of amino acids in plasma samples was analyzed within 65 minutes by HPLC with pre-column derivatization by OPA/3-MPA. This was a gradient RP-HPLC method that was performed using two solvents with a ratio of methanol and sodium acetate. L-norvaline internal standard was used as the reference peak for amino acids. Standard mixture of amino acids was used to determine the concentrations of amino acids.         Results: According to the values of coefficient of variation obtained for each amino acid, the results indicated a good chromatographic separation of amino acids by this method. The use of OPA/3-MPA derivative reagent increased the efficiency and resolution of amino acids chromatographic separation.       Conclusion: Due to simple preparation and accurate assessment, determination of plasma amino acids using OPA/3-MPA derivatives and RP-HPLC is a suitable method in many clinical samples.

RP-HPLC/pre-column derivatization for analysis of omeprazole, tinidazole, doxycycline and clarithromycin.

A validated, reliable and accurate reversed-phase high performance liquid chromatographic method using pre-column derivatization was adopted for the simultaneous determination of two ternary mixtures containing omeprazole, tinidazole and doxycycline hyclate or clarithromycin. Separation was achieved on a C18 column, through a gradient elution system using acetonitrile-methanol-water adjusted to...

Characterization of Jamaican agro-industrial wastes. Part I: characterization of amino acids using HPLC: pre-column derivatization with phenylisothiocyanate.

Jamaican agro-industries generate large quantities of wastes, which are either discarded or under-utilized. In order to evaluate the possible utilization of these wastes, it is necessary that the profiles of the major biochemical groups be developed. This paper describes the determination of the amino acid composition of coffee, citrus, and rum distillery wastes using a reversed-phase high-perf...

Performance of 2-Amino Tetraphenyl Porphyrin as Stationary Phase in RP-HPLC of Amino Acids

The search for new stationary phases has been one of the predominant concerns in high performance liquid chromatography (HPLC) in order to achieve better resolutions, longer column lives, and reduce the time of analysis. A chromatographic packing for separation of underivatized amino acids (AAs) were prepared by dynamically coating 2-amino tetraphenyl prophyrin (atpp) on a C-18 reversed-pha...

HPLC-FLD for the simultaneous determination of primary and secondary amino acids from complex biological sample by pre-column derivatization.

A rapid, sensitive, and reproducible pre-column derivatisation procedure has been established for the simultaneous determination of 20 amino acids by high-performance liquid chromatography using fluorescence detection. The amino acids were derivatized using o-phthalaldehyde and 9-fluorenylmethyl-chloroformate reagents. The optimal conditions for simultaneous separation and detection of both pri...

Automated pre-column derivatization and its application to amino-acid analysis using high-performance liquid chromatography

Although chemical modification of analytes is a well-accepted adjunct to chromatography, little attention had been paid to the automation of pre-column derivatization techniques. An automated pre-column system would offer advantages when analyses are performed using labile derivatives; and also when the time between injections is long and so staff time spent on achieving maximum analytical outp...
