چکیده مقاله:
Background: Based on the reports, high frequency of special alleles of HLA class II genes might be associated with susceptibility to or protective from a particular cancer. These alleles might vary depending on the geographical region. Here we investigate the association between alleles of HLA class II genes and breast cancer in Iranian women. Methods: 100 patients with pathologically proved breast cancer who referred to Cancer Institute, Tehran University of Medical Sciences in Tehran, Iran, were divided to two groups based on ages (40 years old and less/ or more than 40 years old) and were randomly selected and compared with a group of 80 healthy blood donor subjects. HLA class II alleles were determined by amplification of DNA with polymerase chain reaction (PCR) method followed by HLA-typing using sequence-specific primer (SSP) for each allele. Results: The most frequent alleles in the DR and DQ regions in group 1 (40 years old and less) in comparison with control group were HLA-DQA1*0301 (p=0.002) and HLA-DQB1*0302 (p>0.05). In contrast HLA-DQA1*0505 (p=0.004) had significantly lower frequency in this group compared with control group. Patients of group two (more than 40 years old) had a higher frequencies of HLA-DQA1*0301 (p=0.001) and HLA-DRB1*1303 (p=0.02) and a lower frequency of HLA-DQA1*0101 (p=0.002) compared to healthy control. Conclusion: These findings provide information of a positive and negative association between certain alleles of HLA class II and breast cancer in our population and also might support that the pattern of inheritance in the early and late onset of breast cancer differ substantially.

ULCERATIVE COLITIS AND HLA CLASS II PHENOTYPING IN IRAN
Ulcerative colitis (UC) is a nonspecific acute and chronic inflammatory bowel disease that diffusely involves the colonic mucosa. The etiology of UC has not yet been elucidated fully. However, many studies have found that immunologic disorders may play a role in the pathogenesis of UC. In addition, due to an increased frequency of UC in families, especially an increased monozygotic compared...
متن کاملAssociation between HLA-Class I and HLA-Class II Alleles and Mycobacterium Tuberculosis Infection in Iraqi Patients from Baghdad City
Background: Pulmonary tuberculosis (PT) is one of the endemic diseases in Iraq, and among the suggested predisposing factors are alleles of the human leukocyte antigen (HLA) system. We sought to investigate the association between HLA-class I (A and B) and -class II (DR and DQ) alleles in a sample of PT Iraqi patients. Methods: lymphocytes of 105 PT patients and 40 controls were phenotyped for H...
متن کاملبررسی آلو آنتی ژنهای HLA-B، HLA-A و HLA-C در جمعیتی از استان اصفهان
A random sample of 500 healthy unrelated subjects from Esfahan province were HLA typed for A, 8 and C locus antigens. The lymphocytes were separated from 5 ml of whole peripheral blood and were typed for HLA _ A, 8 and C by using the standard two stage microlymphocytotoxicity NIH technique. The findings suggest that in the study population, frequency of HLA-Al' A2, A3, Ag HLA-8s, 83S and HLA-CW...
متن کاملHLA class II regulation and structure. Analysis with HLA-DR3 and HLA-DP point mutants
Point mutations that affect HLA-DR structure or expression have not previously been described. In the present study, we isolated such mutants by immunoselection of an ethyl methanesulfonate-mutagenized HLA-DR3 cell line with an anti-HLA-DR3 monoclonal antibody, 16.23. To facilitate analysis, we used a parent cell line with a preexisting deletion of one haplotype encompassing DR and DQ alpha and...
متن کاملHLA Class I and Class II Genes Distribution of the Sistanis in Iran
Background: The high polymorphism in the human leukocyte antigen (HLA) genes can be used as an identity of individuals to compare with other populations. This extreme polymorphism in the HLA system is accountable for the differences in alleles and haplotypes among ethnic groups, populations, and the inhabitants of many regions. Objective: To define the frequency of HLA alleles and haplotypes am...
متن کاملPhylogenetic Applications of HLA Class II Loci
Human Leukocyte Antigen (HLA) loci widely known for their role in generation of immune responses are often considered to be effective in reconstructing human phylogenies due to high degree of polymorphism and rarity of recombination observed at HLA loci. In the present study, we have made an attempt to substantiate the phylogenetic potential of HLA class II loci by analyzing DRB1, DQA1 and DQB1...
