Tumor Suppressor p53 Can Protect Normal Cells Against Dendrosomal Curcumin-Induced Apoptosis
Authors
Abstract:
Curcumin is a natural substance with anti-cancerous properties without many disadvantages of currently-used anticancer drugs. Its toxicity is significantly higher in tumor cells compared with normal cells. We hypothesized the difference of p53 function between normal and tumor cells as one of the presumable causes of this phenomenon. We knocked down the expression of p53 in normal fibroblasts using anti-p53 siRNA and subsequently explored the effects caused by dendrosomal curcumin- a novel nanoformulation of curcumin- on these cells in terms of apoptosis induction and gene expression analysis. The results of MTT assay demonstrated dendrosomal curcumin is selectively cytotoxic for melanoma cancer cells without any considerable effects on normal fibroblasts. Knocking-down of p53 in normal fibroblast cells caused increase of NF-κB1 and decrease of p21 expression level. Treating p53-suppressed normal fibroblast cells with dendrosomal curcumin led to a robust increase in apoptosis rate of the cells. Taken together, these results imply the fact that p53 can protect normal cells from dendrosomal curcumin-induced apoptosis. Therefore, dendrosomal curcumin- in addition to being a chemotherapeutic compound-represents potential capacities to be used as an effective chemopreventive agent.

full textCurcumin impairs tumor suppressor p53 function in colon cancer cells.
Curcumin (diferuloylmethane) is being considered as a potential chemopreventive agent in humans. In vitro it inhibits transcription by NF-kappaB, and the activity of lipoxygenase or cyclooxygenase enzymes, which facilitate tumor progression. In vivo it is protective in rodent models of chemical carcinogenesis. Curcumin contains an alpha,beta-unsaturated ketone, a reactive chemical substituent t...
full textThe tumor suppressor p53 can both stimulate and inhibit ultraviolet light-induced apoptosis.
We have previously shown that the tumor suppressor p53 can play a protective role against UV-induced apoptosis in human fibroblasts. In the present study, we investigated whether the protective function of p53 expression is established before or after UV irradiation. Using a stable human cell line expressing a murine temperature-sensitive p53 in which p53 function could be tightly and reversibl...

objective: the anti-cancer properties of curcumin, a poliphenol extract from the rhizome of curry, has been confirmed by many investigators. however, low levels of uptake, tissue distribution and rapid metabolism has limited its application as an anti-cancer drug. this study is aimed at increasing curcumin's water solubility due to a biodegradable, neutral and non-toxic micellar nano-carrier ca...
full textThe p53 tumor suppressor protein protects against chemotherapeutic stress and apoptosis in human medulloblastoma cells
Medulloblastoma (MB), a primitive neuroectodermal tumor, is the most common malignant childhood brain tumor and remains incurable in about a third of patients. Currently, survivors carry a significant burden of late treatment effects. The p53 tumor suppressor protein plays a crucial role in influencing cell survival in response to cellular stress and while the p53 pathway is considered a key de...
full textInduced Neural Stem Cells Protect Neuronal Cells against Apoptosis
BACKGROUND Neuronal cells are vulnerable to many stresses that can cause apoptosis. Reprogramming of fibroblasts into induced neural stem cells (iNSCs) is a potentially unlimited source of neurons. Discovering agents that can provide neuronal protection against these apoptotic stimuli is important for developing therapeutic strategies for various brain diseases. MATERIAL AND METHODS We invest...
