Abstract:

Objective(s): Due to wide range of medical applications as bactericidal agents, nanosilver particles (NSPs) are manufactured worldwide in large quantities. However, potential toxicity impacts of NSPs in humans and animals still remain poorly understood. The objective of this study was to investigate clinical observations, mortality and pathological changes in rats following intraperitoneal administration of different doses of NSPs. Methods: In this study, rats were administered intraperitoneally over a period of 5 days with repeated doses at different dose levels (20, 80, 320 mg/kg) of NSPs (20 nm). Rats were euthanized 14 days after the treatment. Animal mortality, clinical sings, food intake and body weight were evaluated. Histopathology was performed on heart, lung, liver and kidneys of experimented animals. Results: There was a significant decrease in the body weight of animals in high dose group following fourteen days of exposure. Also, there was significant decrease in food intake during the treatment period in high dose group. Histological tissue sections indicated that NSPs induced multi-organ pathological lesions including severe alveolar edema, hemorrhage and inflammation in lungs, myocytolysis, congestion and edema in heart, inflammation and congestion in kidney and liver. Conclusions: The results obtained two weeks following intraperitoneal injection indicate that the administration of high doses of NSPs could induce histopathological complications in heart, lung, liver and kidneys in rats. No significant pathological effects were observed in low and intermediate doses. More toxicological investigations are needed in relation of the application of NSPs with their potential threat as a medical tool.

Uptake of Gold Nanoparticles in Several Rat Organs after Intraperitoneal Administration In Vivo: A Fluorescence Study

BACKGROUND The gold nanoparticles (GNPs) have potential applications in cancer diagnosis and therapy. In an attempt to characterise the potential toxicity or hazards of GNPs as a therapeutic or diagnostic tool, the fluorescence spectra in several rat organs in vivo were measured after intraperitoneal administration of GNPs. METHODS The experimental rats were divided into control and six group...

Effect of pentoxifylline on histomorphological changes of kidney after Ecstasy (MDMA) administration in male wistar rat.

Methylenedioxymethamphetamine (MDMA),the Ecstasy brand, leads to cell death in many tissues such askidney because of its oxidative properties. The present study aimed to investigate the possible effects ofpentoxifylline as a vasodilators on ecstasy induced renal damage. This experimental study was carried out infour groups of six male wistar rats weighing 250-300 g (n=24). The control group was...

Blood toxicity of silver nanoparticles in Pregnant Wistar Rats

Background: Investigation of toxicity of silver nanoparticle, especxially blood toxicity, is necessary because this nanoparticle is used a lot in diffrerent parts of life and invironment. The aime of this is evaluation the effects of silver nanoparticles with different concentrations on blood cells pregnant Wistar rats. Materials and Methods: In this case control study, 30 pregnant rat wer...

Evaluation of Clarithromycin Pharmacokinetics after Single and Repeated oral Administration of Atorvastatin in Hyperlipidemic Wistar Rats

To evaluate the clarithromycin pharmacokinetics after atorvastatin single and repeated dose administration in induced hyperlipidemic Wistar rats. A standard cholesterol diet (2% cholesterol) was used to induce hyperlipidemia and feed was given to rats for 6-8 weeks. Group’s allocation as follows: G1-G3: Induced hyperlipidemic rats pretreatment of atorvastatin as a single dose followed by clarit...
