The transcription of Hypoxia-inducible factors (hif-1, hif-2) during normal development of Beluga sturgeon (Huso huso Linnaeus, 1758)
Authors
Abstract:
In the present study, the levels of the hypoxia-inducible factors (hif-1, hif-2) transcripts were evaluated during development of an ancient fish species, Beluga sturgeon (Huso huso) in a normal oxygen environment. The transcripts of hif-1, hif-2 were detected in all developmental time-points of H. huso from embryos to juvenile fish. Changes in normalized hif-1, hif-2 mRNA expression by using the RPL6 over development was statistically significant over time (P < 0.05). Our results revealed that the mRNA expression of hifs was low during embryonic development and then upregulated significantly in the time of hatch and early larval time-points whereas, in the late larval development and juvenile stage started to decline. The upregulation of hif-1, hif-2 transcripts at the time of hatch and during yolk sac fry development of H. huso is likely tied to the role of hifs in organogenesis and angiogenesis.

Semen obtained from four Beluga males (Huso huso) was cryopreserved using extender; Tris-sucroseKCl (30mM Tris, 23.4mM sucrose, 0.25mM KCl, PH 8.0) supplemented with MeOH at concentration of 5%, 10% and 20%. Semen was diluted, respectively, with ratios of 1:0.5, 1:1 and 1:2 with extender and frozen in liquid nitrogen vapor.Frozen sperms after 30 and 60 days were excluded from freezing. Experime...
full textPodocyte expression of hypoxia-inducible factor (HIF)-1 and HIF-2 during glomerular development.
The heterodimeric transcription factors, hypoxia-inducible factor (HIF)-1 and HIF-2, are essential for the maintenance of cellular oxygen homeostasis. In response to hypoxia, stabilized HIF-1alpha and HIF-2alpha proteins bind HIF-1beta and initiate expression of genes that alleviate hypoxic stress, including those promoting neovascularization. Both HIF-1 and HIF-2 stimulate transcription of vas...
full textEffect of freezing rate on motility, adenosine triphosphate content and fertilizability in beluga sturgeon (Huso huso) spermatozoa.
Broodstock selection programs are currently underway for sturgeon species. To complement and further these selection programs we need to develop sperm cryopreservation procedures. In the present study, we describe the effects of freezing rate (-10°C, -15°C, -20°C, -30°C and -40°C/min) on gamete quality characteristics (i.e., duration of motility (s), motility percentage (%), ATP content (nmol/1...

In this study, the effects of sublethal concentrations of the Endosulfan on gene expression of hypoxia-inducible factor (HIF-1 α, HIF-2 α) and vascular endothelial growth factor (VEGF) in the gill tissue of Persian sturgeon fingerling (Acipenser persicus) were studied. full textPredominant Role of Hypoxia-Inducible Transcription Factor (Hif)-1 versus Hif-2 in Regulation of the Transcriptional Response to Hypoxia
Tumor hypoxia induces the up-regulation of a gene program associated with angiogenesis, glycolysis, adaptation to pH, and apoptosis via the hypoxia-inducible transcription factors (Hifs) 1 and 2. Disruption of this pathway has been proposed as a cancer therapy. Here, we use short interfering RNAs to compare specific inactivation of Hif-1 or Hif-2 and show markedly different cell type-specific e...
