SYNTHESIS OF 4,4'-BIS-PYRIMIDINES AND SOME RELATED BIS-FUSED PYRIMIDINES

Authors: not saved
Abstract:

2,2'-Disubstitutedamino-4-4'-bis-pyrimidines (3a-m) have been prepared by linking 2,4-dichloro-6-methyl-5-nitropyrimidine (1) with several diamines and subsequent reaction with suitable amines. Some of these compounds (3c and 3j) which were reduced catalitically, on treatment with nitrous acid, urea in pyridine, and boiling thionyl chloride gave the corresponding bistriazolopyrimidin (6), bis-purines (7), and bis-isothiazolopyrimidines (8). Two bis-imidazo[l,2-a] pyrimidines (9 and 10) have also been prepared

Synthesis of new fused pyrimidines by isocyanate and isothiocyanate.

o-Aminonitrile or o-aminoester compounds were cyclized to fused pyrimidines by reacting with ethyl iso(thio)cyanatoacetate in pyridine, and then were methylated, halogenated and subsequently displaced by the amines studied.

Synthesis of Fused Pyrimidines from Nitroarenes via ortho-Nitroaromatic Carboximes

A synthesis of functionalized fused pyrimidine derivatives from the respective ortho-nitroaromatic carboximes is described. Commercially available nitroarenes were transformed according to a known manner into the ortho-nitroaromatic carboximes. Selective catalytic reduction of the nitro group in these compounds followed by condensation of the amines formed with various orthoesters, resulted in ...

Microwave assisted synthesis of bis and tris(ω-bromoacetophenones): versatile precursors for novel bis(imidazo[1,2-a]pyridines), bis(imidazo[1,2-a]pyrimidines) and their tris-analogs

BACKGROUND α-Bromination of the side chain of aromatic ketones using NBS in the presence of p-toluenesulfonic acid (p-TsOH) in acetonitrile is very common. However, regioselective bromination of bis and tris(ω-bromoacetophenones) with NBS in the presence of p-TsOH in acetonitrile under microwave irradiation is quite novel. The bis- and tris(ω-bromoacetophenones) are used in synthesis of bis and...

Expanding the scope of fused pyrimidines as kinase inhibitor scaffolds: synthesis and modification of pyrido[3,4-d]pyrimidines.

Fused pyrimidine cores are privileged kinase scaffolds, yet few examples of the 2-amino-pyrido[3,4-d]pyrimidine chemotype have been disclosed in the context of kinase inhibitor programs. Furthermore, no general synthetic route has been reported to access 2-amino-pyrido[3,4-d]pyrimidine derivatives. Here we report a versatile and efficient chemical approach to this class of molecules. Our strate...

Synthesis and anti-histaminic activity of some novel pyrimidines.

Novel pyrimidines were prepared by the condensation of chalcones of 4'-piperazine acetophenone with guanidine HCl. The structures of the synthesized compounds RP 1-5 were assigned on the basis of elemental analysis, IR, (1)H NMR and mass spectroscopy. These compounds were also screened for anti-histaminic activity. The recorded percentage of histamine inhibition showed a significant anti-histam...
