Abstract:

In the present study, 2, 4-dichlorophenoxyacetic acid (2, 4-D) was assessed individually for its effectiveness to induce somatic embryogenesis in tea (Camellia sinenesis L.). Embryonic axes and cotyledons explants were dissected from the seeds. Explants were cultured on Murashige and Skoog (MS) medium containing 0, 1 and 5 mM 2, 4-D alone for embryonic axes and 0, 1 and 5 mM 2, 4-D along with 0 and 0.5 mM IBA for cotyledons. Embryos were observed in embryonic axes explants cultured on MS medium containing 1mM 2, 4-D. No somatic embryos were seen on cotyledons explants.

Somatic embryogenesis and plant regeneration from zygotic embryo explants of onion

The study was undertaken to examine the effect of two synthetic auxins on somatic embryogenesis and plant regeneration from mature embryo explants of 16 onion cultivars. Cotyledons were removed from the embryos and remaining portions were cultured on MS medium supplemented with 1.0 mg/L 2,4-D and 5.0 mg/L picloram alone or in combination (1.0 mg/L 2,4-D + 2.5 mg/L picloram) to produce embryogen...

Somatic embryogenesis and plant regeneration in Gloriosa L.

Friable callus was initiated from shoot apices of Gloriosa superba L. on basal MS medium supplemented with 2, 4-D (4mg L(-1)) + Kn(5 mg L(-1)) + CH(10 mg L(-1)) + CW(20%). Subculture of callus on the same medium after 4-5 weeks showed induction of large number of somatic embryos, which was confirmed with histological studies. Development of embryoids in plantlet took place when the embryogenic ...

Somatic Embryogenesis and Plant Regeneration in Evening Primrose (Oenothera biennis L.)

Callus induction responses and regeneration through callus-mediated SE were studied from evening primrose (Oenothera biennis L.) as an important medicinal plant of Onagraceae mainly known for its gamma-linoleic acid (GLA) content. The effects of cytokinins, 6-Benzylaminopurine (BAP) and N-(2-furfurylamino)1-H-purine-6-amine [Kinetin (Kin)] and their concentrations (0, 0.5, 1.0 and 1.5 mg/l) in ...

Somatic Embryogenesis and Plant Regeneration in Cucumber

The embryogenic capacity of seven cucumber (Cucumis sativus L.) cultivars was examined by tissue culture of cotyledon, young first-leaf, and internode explants. Somatic embryogenesis frequencies differed significantly among the tested cultivars, and ‘Fushinarimidori’ produced the highest number of embryos from either cotyledons or young first leaves. Cotyledonand first-leaf-derived calluses pro...

Induction of Callus and Somatic Embryogenesis from Cotyledon Explants of Fagonia indica Burm.

To explore the potential for in vitro rapid regeneration of Fagonia indica Burm., cotyledon explants were obtained from 10-day old aseptically germinated seedlings that cultured on MS medium which supplemented with 1.0-5.0 µM 2,4-D. Callus was initiated from cotyledon explant on MS medium supplemented in all concentrations of 2,4-D investigated. 2,4-D was more effective than IAA and NAA for cal...
