Research Paper: Optimization of Transient Focal Cerebral Ischemia Model by Middle Cerebral Artery Occlusion
Authors
Abstract:
Introduction: Cerebral ischemia is one of the most common causes of death in human populations in the industrial communities. The need for animal models is inevitable to study the pathophysiology and treatment of cerebral ischemia in human. The current study aimed at evaluating the strengths and weaknesses of different techniques used to create ischemia in previous studies and optimizing the transient focal cerebral ischemia model by Middle Cerebral Artery Occlusion (MCAO). Methods: In the current study, 35 male Wistar rats were divided into 4 groups. The weight of rats in the groups 1 and 2 ranged 250 to 270 g with ischemic induction duration of 60 and 90 minutes, respectively. The weight of rats in the groups 3 and 4 ranged 250 to 310 g with ischemic induction duration of 60 and 90 minutes, respectively. The ischemia was performed by the MCAO method using a special filament. Motor neurological disorders were assessed 24 hours after ischemia. The ischemic area was appeared using Triphenyl-Tetrazolium Chloride (TTC) staining. Histopathological examination of the ischemic area was performed using the Nissl staining. Results: In a similar duration of ischemia, the ischemic lesion volume was significantly greater in the group with lower weight. The mortality rate was lower in the groups with lower weights compared with other groups. Conclusion: Based on the results of the present study, it is better to use rats with lower weights to create transient focal cerebral ischemia model using MCAO method.

Focal Cerebral Ischemia Model by Endovascular Suture Occlusion of the Middle Cerebral Artery in the Rat
Stroke is the leading cause of disability and the third leading cause of death in adults worldwide. In human stroke, there exists a highly variable clinical state; in the development of animal models of focal ischemia, however, achieving reproducibility of experimentally induced infarct volume is essential. The rat is a widely used animal model for stroke due to its relatively low animal husban...
full textBiochemical evaluation of focal non-reperfusion cerebral ischemia by middle cerebral artery occlusion in rats.
Cerebral ischemia is an important event in clinical and surgical neurological practice since it is one of the diseases that most compromise the human species. In the present study 40 adult rats were submitted to periods of focal ischemia of 30, 60 and 90 min without reperfusion and animals submitted to a sham procedure were used as controls. We analyzed the levels of ATP, malondialdehyde and ca...
full textNeuroprotective effect of curcumin in middle cerebral artery occlusion induced focal cerebral ischemia in rats.
Free radical induced neuronal damage is implicated in cerebral ischemia reperfusion (IR) injury and antioxidants are reported to have neuroprotective activity. Several in vitro and in vivo studies have proved the antioxidant potential of curcumin and its metabolites. Hence, in the present study the neuroprotective potential of curcumin was investigated in middle cerebral artery occlusion (MCAO)...
full textEffect of pentoxifylline on brain edema in a rat model of transient focal cerebral ischemia
Pervious studies have shown that pentoxifylline (PTX) has beneficial effects in reduction of stroke and brain trauma injuries in experimental animals. However, there is very little and controversial information about the effect of PTX on brain edema in cerebral ischemia. Therefore, the aim of this study was to determine the effects of different doses of PTX on brain edema and neurological m...
full textEffect of Combination Therapy Using Hypothermia and Granulocyte Colony-Stimulating Factor in a Rat Transient Middle Cerebral Artery Occlusion Model
Background: Stroke is the third leading cause of death. Hypothermia has been recognized as an effective method in reducing brain injury. In this study, we assessed the effects of granulocyte colony-stimulating factor (G-CSF) as a neuroprotective agent and mild hypothermia on mortality, behavioral function, infarct volume, and brain edema in Wistar rats. Methods: Forty male rats were used in fiv...
full textNeuroprotective properties of the novel antiepileptic drug levetiracetam in the rat middle cerebral artery occlusion model of focal cerebral ischemia
Levetiracetam (LEV) is a new antiepileptic drug with a promising preclinical profile involving both anticonvulsant and antiepileptogenic effects in kindling models. The latter stimulated the present study to compare its neuroprotective properties with the potent and selective, non-competitive NMDA antagonist, MK-801, in the rat middle cerebral artery occlusion model. Twenty-four hours after a t...
