RAPD Analysis for Sex Determination in Pistacia vera L.
Authors
Abstract:
Random Amplified Polymorphic DNA (RAPD) analysis of Pistacia vera L. was used to distinguishing sex type in progeny plants. Varied genotypes of the pistachio were analyzed by PCR amplification of a ten base primer previously found to be linked to the expression of a 945bp amplification band in female trees and its absence in males. Results revealed that genetic material from cultivar samples amplified well with relative primer, but some male progeny produced the 945bp band when amplified with the 10bp primer. Despite numerous subsequent tests for reamplification of the DNA, unfortunately, these male progeny failed to give reproducible results from its sample. None the less, presence of the 945bp amplification band in a male sample proved that an error rate of 15% is possible in sex determination of P. vera by RAPD analysis with the 10bp primer.

application of rapd (random amplified polymorphic dna) marker for sex determination of pistacia vera l.
sex determination in pistacia as a dioecious plant is economically desirable. identification of the male and female plants has a great value for pistacia yield production. in the present study, leaf samples from four female plants from cultivars of pistacia including akbary, akbar aghaei, fandoghi, and kalleh ghochi as well as male plants were used for rapd pcr amplification. among 32 primers w...
full textDetermination and assesment of the sex chromosomes of male trees of pistachio (Pistacia vera L.) using in vitro culture
The chromosome number and karyotype analysis of male trees of Pistacia vera L. was investigated to determine the sex chromosomes. For karyological determination, root tips were obtained from in vitro derived shoots. Afterwards fifty plates of mitosis were used for the karyotyping, the five best metaphase plates were measured for karyotype analysis, karyogram and idiogram. The conclusions obtain...

The effect of different air temperatures (40, 50, 60 and 70°C), air velocities (1 and 2 m/s) and bed depth (monolayer, 3 and 5 cm) on drying kinetics and lipid quality (peroxide value and free fatty acids) of a variety of Persian Pistachio nuts (Abasali variety) were studied. Additionally, effective diffusivity (D ) was determined eff by using Fick’s second law varied from 7.061×10 to 2.105×10 ...
full textDynamics of growth and phytomass allocation in seedlings of Pistacia atlantica desf. versus Pistacia vera L. under salt stress
Salinity represents a major environmental constraint which can severely limit plant establishment in the arid and semi-arid areas in Tunisia. The present work was carried out to study the response of Pistacia vera L. and Pistacia atlantica Desf. seedlings to salt stress. Evaluation of salinity effects on both species was investigated using five increasing NaCl concentrations (0, 20, 40, 60 and ...
full textThe Anti-Inflammatory and Antioxidant Potential of Pistachios (Pistacia vera L.) In Vitro and In Vivo
Several reports have demonstrated the effectiveness of pistachio against oxidative stress and inflammation. In this study, we investigate if polyphenols extracts from natural raw shelled pistachios (NP) or roasted salted pistachio (RP) kernels have anti-inflammatory and antioxidant properties at lower doses than reported previously, in both in vitro and in vivo models. The monocyte/macrophage c...
full textAlternate bearing and the possible role of carbohydrates in bud abscission of pistachio (Pistacia vera L.)
Pistachio produces an abundant number of inflorescent buds every year (during spring and early summer) which abscise mainly on fruiting branches during seed development. Competition of the developing ovaries/seeds with inflorescent buds for nutrients was suggested as being primarily responsible for bud abscission. Although leaf N correlated with bud retention, foliar application of N did not re...
