Producing Soluble Human Programmed Cell Death Protein-1: A Natural supporter for CD4+T cell Cytotoxicity and Tumor Cells Apoptosis
Authors
Abstract:
Background: Programmed cell death protein-1 (PD-1)/PD-L1 pathway is one of the immune checkpoint pathways involved in regulation of the immune responses and suppression of anti-tumor defense. PD-1/B7-H1-blocking antibodies improve immune responses such as cytotoxic activity of CD8+/CD4+T cells and also increase mortality of tumor cells; however their use is accompanied by adverse effects in patients. Objectives: We aimed to produce a native blocker of human PD-1/PD-L1, for developing T cells cytotoxicity and tumor cells apoptosis. Materials and Methods: We designed and cloned soluble human PD-1-GFP-pcDNA3.1/hygro construct in Escherichia coli strain TOP10 cells and then transfected this construct into HEK cells. Concentration of secreted shPD-1 in the supernatant was measured and the supernatant was used for blocking PD-L1 on the MDA-MB-231 cells. The cytotoxicity of CD8+/CD4+T cells and apoptosis of MDA-MB-231 cells under the influence of shPD-1 in the co-culture of T cells with MDA-MB-231 cells were evaluated using flow cytometry technique. Results: GFP expression in the transfected cells illustrated the successful designing, transfection and production of shPD-1. Soluble human PD-1 concentration in supernatant of transfected HEK cells was significantly higher than untransfected cells. In addition, shPD-1 significantly blocked PD-L1 on the MDA-MB-231 cells, improved cytotoxicity of CD4+T cells and increased apoptosis of MDA-MB-231 cells. Conclusions: Overall, increased CD4+T cell cytotoxicity and tumor cells apoptosis under the influence of shPD-1, confirmed effectiveness of shPD-1 as a natural blocker of PD-L1and as an augmenter of the anti-tumor immune responses.

Apoptosis: programmed cell death.
C urrently there is much interest and excitement in the understanding of how cells undergo the process of apoptosis or programmed cell death. Understanding how, why, and when cells are instructed to die may provide insight into the aging process, autoimmune syndromes, degenerative diseases, and malignant transformation. This review focuses on the development of apoptosis and describes the proce...
full text[Apoptosis--programmed cell death].
Apoptosis is a highly organized physiologic mechanism of destroying injured and abnormal cells as well as maintaining homeostasis in the multicellular organism. Apoptosis is tightly controlled at the genetic level and dysregulation of the apoptosis machinery may contribute to a variety of diseases. Pharmacologic manipulation of this pathway is a novel therapeutic target.
full textApoptosis: a review of programmed cell death.
The process of programmed cell death, or apoptosis, is generally characterized by distinct morphological characteristics and energy-dependent biochemical mechanisms. Apoptosis is considered a vital component of various processes including normal cell turnover, proper development and functioning of the immune system, hormone-dependent atrophy, embryonic development and chemical-induced cell deat...
full textInduction of apoptosis in human tumor cell lines by platelets
Introduction: It has been reported that platelets can eradicate tumor cells in vitro, although the mechanism of this effect has not been determined. The effect of platelets on the induction of apoptosis in tumor cells is largely unknown. Materials and methods: To investigate this effect, two human hematologic cell lines, K562 and Daudi, were independently faced with unstimulated and thromb...
full textProgrammed Cell Death (Apoptosis) and Cancer Chemotherapy.
BACKGROUND: Programmed cell death involves a genetic reprogramming of the cell to promote an energy-dependent cascade of biochemical and morphological changes within the cell that result in its death and elimination. METHODS: The regulations and mechanisms of programmed cell death are reviewed with an emphasis on how derangement of this mechanism may be involved in modulating responsiveness to ...
full textEffect of Clarithromycin on Enhancement of Doxorubicin-induced Cytotoxicity and Apoptosis in A549 Tumor-Cell Line
Background and purpose: Nowadays, multi-drug regimens or combination therapy are new approaches in cancer treatment that combine two or several therapeutic agents. Combination therapy aims at improving the therapeutic efficacy of anticancer drugs while reducing toxicity and preventing drug resistance. The aim of this study was to determine the synergistic effect of combined clarithromycin and d...
