PLLA/HA Nano composite scaffolds for stem cell proliferation and differentiation in tissue engineering
Authors
Abstract:
Abstract Due to their mulitpotency, Mesenchymal stem cells (MSCs), have the ability to proliferate and differentiate into multiple mesodermal tissues. The aim of this study was to isolate MSCs from human Umbilical Cord (hUCMSCs) to determine their osteogenic potential on nanofibrous scaffolds. To this end, Poly (L-lactic acid) (PLLA)/Nano hydroxyapatite (HA) composite nanofibrous scaffolds were prepared by electrospinning. The structure and morphology of the scaffolds were investigated using scanning electron microscopy. Human mesenchymal stem cells (MSCs) were isolated from the umbilical cords and cultured in the PLLA/HA scaffold. The viability and proliferation of the cells was then determined by an MTT assay. Cellular adhesion, proliferation and osteogenic differentiation were assessed in these constructs using a range of histological and microscopic techniques. The osteogenesis assays indicated the superiority of nanofibrous scaffolds in supporting MSCs undergoing bone differentiation. Collectively, the bone construct prepared with PLLA/HA scaffold and proliferated MSCs would be a suitable candidate for use in bone regenerative medicine. Keywords: hUCMSCs; nanofibrous scaffolds; hydroxyapatite; PLLA; Cell proliferation; Bone differentiation.

Bones constructed by tissue engineering are being considered as valuable materials to be used for regeneration of large defects in natural bone. In an attempt to prepare a new bone construct, in this study, proliferation and bone differentiation of marrow-derived mesenchymal stem cells (MSCs) on our recently developed composite scaffolds of nano-, micro-hydroxyapatite/ poly(l-lactic acid) were ...
full textStem Cell Scaffolds for Tissue Engineering
Tissue engineering is the science that combines the principles of cellular biology, material science and biomedical engineering techniques in order to obtain biological substitutes for regenerating, replacing, modifying, repairing or restoring the function of organs and tissues. For stem cells to be used in tissue engineering a scaffold is essential to provide the necessary support for the tran...

Human adipose tissue represents an abundant, practical and appealing source of donor tissue for autologous cell replacement. Recent findings have shown that stem cells within the stromalvascular fraction of adipose tissue display a multilineage developmental potential. Adipose tissue-derived stem cells can be differentiated towards adipogenic, osteogenic, chondrogenic, myogenic and neurogenic li...

Biomaterial and scaffold development underpins the advancement of tissue engineering. Traditional scaffolds based on biodegradable polymers such as poly(lactic acid) and poly(lactic acidco-glycolic acid) are weak and non-osteoconductive. For bone tissue engineering, polymer-based composite scaffolds containing bioceramics such as hydroxyapatite can be produced and used. The bioceramics can be e...

Tissue engineering may be defined as the application of biological, chemical and engineering principles toward the repair, restoration or regeneration of living tissue using biomaterials, cells and biologically active molecules alone or in combinations. The rapid restoration of tissue biomechanical function represents a great challenge, highlighting the need to mimic tissue structure and mechan...
