Abstract:

In this study Escherichia coli DE3 containing expression vector (pET21a) with cloned Persian sturgeon growth hormone (psGH) gene was grown in 10 mL LB broth on a 150 rpm shaker, at the temperature of 37 °C. At the late log phase (determined by OD standard curve) 100 &muL isopropyl &beta-D-1-thiogalactopyranoside (IPTG) was added for induction of GH synthesis. Samples were taken every 2 hours and after bacterial cells lysis crude extracts with recombinant proteins inclusion bodies (IB) were loaded on 15% SDS-PAGE gel. Thenafter staining, comparative concentrations of rpsGH were measured by densitometric scanning of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) gel  it was more than 90 %. The maximum yield of GH was observed after 4 hours of growth. To recover active psGH from inclusion bodies we used imidozole to obtain most of the total recombinant protein in the soluble fraction. Purification of 6xhisN tag recombinant psGH has been performed using affinity chromatography where nickel was bound to an agarose bead by chelation using NTA (nitrilotriacetic acid) beads. The overall yield of the purified monomeric psGH was approximately 50% of the initial IB proteins. The purification manipulations including IB isolation and solubilisation, protein refolding by dialyze and affinity chromatography ensure yields of biologically active psGH up to 30%. This study shows that, the affinity chromatography is a powerful and very specific method for recombinant proteins purification of  psGH.

Purification and characteristics of porcine growth hormone.

A simple procedure is described for the isolation of porcine growth hormone from the filtrate remaining after the adsorption of adrenocorticotropic hormone (ACTH) onto oxidized cellulose during the commercial manufacture of ACTH. Growth hormone is purified by ion exchange chromatography on diethylaminoethylcellulose. The purified hormone has a specific biological activity nearly twice that of t...

Morphological and Histological Study of Kidney in Juvenile Great Sturgeon (Huso huso) and Persian Sturgeon (Acipenser persicus)

Sturgeons are ancient and commercial fishes which posses’ primitive characteristics and due to their great inflections are able to live in both freshwater and marine environments. So their kidney has great ability for adaptation process. In order to find the structure and distribution of kidney nephrons in juvenile Huso huso and Acipensr persicus, sampling of head, body and caudal parts of kidn...

Expression of growth hormone gene during early development of Siberian sturgeon (Acipenser baerii)

The mRNA expression of growth hormone (GH) gene in early development stages of Siberian sturgeon was investigated using RT-PCR method. Samples were collected from unfertilized eggs up to 50 days post hatched (dph) larvae in 11 different times. Ribosomal protein L6 (RPL6) transcripts were used as the internal standard during quantification of GH mRNA expression. The results showed that the GH mR...

Effects of Daphnia magna and Artemia nauplii on growth performance in Persian sturgeon Acipenser persicus larvae

This study was carried out in a completely randomized design 3×3×4 factorial arrangement in three periods of rearing in spring 1997. Density of 300 larvae was assigned for each oval tank with 0.1m2 surface area, 30cm deep, a volume of 20lit and 0.25 lit/min of water flow per minute at Shahid Rajaie fish breeding and culture center in Sari, north of Iran. The fries were divided into ...
