Abstract:

Background: In mammalian spermatozoa, motility is one of the most important parameters for evaluating the quality of the semen. From a functional point of view, motility is strongly related to the spermatozoa’s ability to manage its energy status, Thus, modulation of energy status could be translated into significant changes in motility patterns. The aim of the present study was to evaluate xylose sugar different levels, on buffalo epididymal sperm motility patterns with computer-assisted sperm analyzers (CASA). Materials and Methods: For this study buffalo bull testicles (15 pairs) were picked up in the march till September 2012 in Urmia local slaughterhouse and transported to the laboratory in a cool container (filled with 5oc ice pack). After adding 5 levels of xylose (1-3-5-10-15 mM), in TCM199 media with 10 percent BSA and ,24 hours incubation in 37ºC was performed. Sperm motility was examined at 1, 3, 5, 7, 10, 12 and 24 hours after incubation with CASA. After collection of data from CASA analysis, Statistical analyses were performed with procedures available in ANOVA of SPSS version 20. Results: The results showed that CASA parameters; rapid progressive motility (Class A, %), progressive motility (Class B, %), motile sperms (Class A+B+C, %), straight line velocity (VSL, μm/s), average path velocity (VAP, μm/s), curvilinear velocity (VCL, μm/s), amplitude of lateral head displacement (ALH, μm), angular displacement (MAD, D),beat cross frequency (BCF, Hz) were significantly higher in control than xylose levels specially in 24 hours after incubation (p<0.05). Conclusion: In conclusion, our results indicate that xylose, at concentrations from 1 to 15 mM, decrease CASA parameters, maybe xylose don’t activate the glycolytic pathway and ATP production at this concentrations in TCM 199 media in 37 ºC.

Cryopreservation of in situ cool stored buffalo (Bubalus bubalis) epididymal sperm

The objectives of this study were to investigate the effects of testis-epididymis cool storage on viability and progressive motility of buffalo epididymal sperm (EP) and to compare the influences of two basic semen extenders on post thaw EP viability and progressive motility. Abattoir collected buffalo testicles were allotted to three storage times (0 h: n = 10; 24 h: n = 10 and 48 h: n = 12). ...

P-103: Effects of In Vitro Selenium Supplementation on Buffalo (Bubalus bubalis) Sperm Quality following Cryopreservation

Background: Artificial insemination is a valuable tool in genetic improvement programs and widely used breeding technique in farm animals. Semen processing and cryopreservation causes considerable damage to sperm DNA, motility apparatus, plasma membrane, acrosomal cap and thus, reduced fertility. Selenium is an integral part of glutathione peroxidase (GSH-Px), an enzyme which protects cell inte...

The in vitro effect of leptin on semen quality of water buffalo (Bubalus bubalis) bulls

The purpose of this study was to evaluate the probable effects of leptin addition in different levels to the semen extender on sperm quality (motility and motility parameters, viability, sperm membrane integrity, and DNA damage). Semen specimens were evaluated immediately after leptin addition, equilibration time and after thawing the frozen semen. Five healthy buffalo bulls (5 ejaculates from ...

Frequency of Toxoplasmosis in Water Buffalo (Bubalus bubalis) in Trinidad

Toxoplasmosis has been reported to occur in several animals and humans causing different clinical manifestations. The study was conducted to determine the frequency of Toxoplasma gondii antibodies (IgG) in water buffalo (Bubalus bubalis) across farms in Trinidad using a latex agglutination test. Of a total of 333 water buffalo tested, 26 (7.8%) were seropositive for T. gondii antibodies. Seropo...

EFFECT OF Zn SUPPLEMENTATION (ZnSO4) ON SPERM MORPHOMETRY OF MURRAH BUFFALO BULLS (BUBALUS BUBALIS)

The objective of the present study was to study the effect of zinc supplementation (ZnSO4) on sperm morphometry of the Murrah buffalo bulls. Eight apparently healthy, sexually mature and clinically normal Murrah buffalo bulls of similar body weight and age group (nearly 3.0 to 6.5 years) were randomly divided into two groups: control and ING groups. The Diet for both the groups was the same exc...
