Abstract:

Background Cryopreservation procedure for poultry semen is not reliable due to structural and biochemical damages which can lead to reduction in sperm quality and fertility. Development of cryopreservation medium has crucial effects for successful recovery of sperm after freezing. The objective of this study was assessment of cryopreservation of rooster semen using hand-made cryopreservation media supplemented with different cryoprotectants. MaterialsAndMethods Two hand-made diluents (Lake and Beltsville media) were assessed for cryopreservation of rooster semen. Moreover, glycerol (3%) and Dimethylacetamide (6%) were applied as cryoprotectants in the diluents for rooster sperm freezing. Experimental treatments were consisting of A: lake with 3% glycerol, B: Lake with 6% DMA, C: Beltsville with 3% glycerol and D: Beltsville with 6% DMA. For qualification of treatments, motion characteristics and viability were measured using computer semen analysis and Eosin- Nigrosi, respectively. Statistical analysis was applied using SPSS software. The average was compared with Tokay Test. Results In a general point of view, the highest significant percentage of sperm with motility and viability were obtained in Lake solution. In Lake solution, glycerol had the higher significant effect for motility and viability compare to DMA. In Beltsevil extender, there were not significant effects between glycerol and DMA. Conclusion From our results, it can be concluded that Rooster semen could be more efficient after freezing-thawing when it cryopreserved with Lake Solution supplemented with glycerol.
