P-188: Mammary Gland Changes during Chlorpromazine Treatment in Female Rats

Authors

Abstract:

Background: Antipsychotic drugs are medications used to treat schizophrenia and other psychotic disorders. Antipsychotic drugs can cause unwanted side effects. Hyperprolactinemia is an important but neglected adverse effect. The aim of the this study was to consider the effects of Chlropromazine -induced hyperprolactinaemia on mammary glands of female rats . Materials and Methods: In this study 32 adult female wistar rats (160 ± 5 g) were randomly divided into four groups (n=8 each) with one group serving as control sham((5 mL/kg of 0.5% methylcellulose solution.), in the drug treated groups female rats were orally administered CPZ (3, 10, 30 mg/kg) once daily for 28 days .The test compound was suspended in 0.5% methylcellulose solution . On day 29th all rats were sacrificed by CO2 inhalation and blood samples were collected for biochemicals analyses. The mammary glands were removed surgically and prepared for histological study. Results: This study showed that treated by CPZ had significant increase in prolactin level in plasma . In the mammary gland, acinous hyperplasia with an increased secretion was observed in females treated with CPZ. Conclusion: These results showed that CPZ-induced hyperprolactinaemia negatively affects on mammary glands and there were mammary gland changes and unexpected breast development.

Changes in lipogenesis during mammary-gland development.

The mammary gland is an excellent system with which to study differentiation. During pregnancy, differentiation can be monitored by measuring the biochemical changes that lead to the unique ability of the lobulo-alveolar tissue to synthesize milk components such as milk fat. Unlike most mammalian tissues, the lactating mammary gland of many species synthesizes medium-chain (C, : o-Clz : o ) fat...

P-58: Evaluation of Ovarian Toxicity Effects of Chlorpromazine in Female Rats

Background: Elevations in serum prolactin levels (hyperprolactinaemia) are a common side effect of conventional antipsychotic treatments. The main objective of this study was to consider the effects of the antipsychotic drug chlorpromazine (CPZ) in female rats reproductive system and to examine the relationship between the effects on reproductive organs and hormone concentrations. Materials and...

The Effects of Chlorpromazine on Reproductive System and Function in Female Rats

Background Chlorpromazine (CPZ), an antipsychotic drug, is associated with increased risk of sexual dysfunction through increasing prolactin levels. The current study evaluates the effect of CPZ-induced hyperprolactinemia on ovarian follicular growth, gonadotropins, and alteration of ovarian source hormones. MaterialsAndMethods In this experimental study, animals were divided into four groups, ...

Changes in Mammary Gland Morphology and Breast Cancer Risk in Rats

Studies in rodent models of breast cancer show that exposures to dietary/hormonal factors during the in utero and pubertal periods, when the mammary gland undergoes extensive modeling and re-modeling, alter susceptibility to carcinogen-induced mammary tumors. Similar findings have been described in humans: for example, high birthweight increases later risk of developing breast cancer, and dieta...
