Abstract:

Background During mammalian spermatogenesis unique and dynamic epigenetic events occur leading to chromatin condensation. Through these events, histone demethylases such as JMJD1A play important roles in compaction of sperm chromatin, due to regulation of histone methylation dynamics and alteration of chromatin structure. As histone methylation is one of the best-characterized modifications in the study of germ cell development, evaluation of presence/ absence of JMJD1A protein in impaired spermiogenesis was aimed in this study. MaterialsAndMethods For this respect, based on spermogram and presence/absence of sperm in testicular sperm extraction (TESE) process, testis tissue samples of infertile men referred to Royan Institute were collected as two groups of TESE negative (n=9) and TESE positive (n=3), respectively. Immunohistochemical analysis of paraffin embedded tissue samples was performed qualitatively, using anti-JMJD1A antibody to elucidate presence/ absence of JMJD1A in nucleus of germ cells. Results Immunohistochemical analysis data showed absence of JMJD1A protein in nucleus of germ cells in TESE negative group compared to TESE positive group.n Conclusion It can be concluded that there is an obvious association between absence of histone demethylation as a chromatin condensing state with impairment of spermatogenesis and male infertility.

P-202: Reduced Expression of JMJD1A Histone Demethylase Gene in Testis Tissues of Infertile Men Referred to Royan Institute

Background: Epigenetic modifications are involved in different cellular processes through regulating chromatin dynamics. histone methylation is an important modification that can be dynamically regulated by histone methyltransferase and histone demethylase enzymes. JMJD1A (also known as JHDM2A and KDM3A) is a histone demethylase specific for H3K9me2/me1. JMJD1A is a key epigenetic regulator tha...

Investigation of Histone Lysine-Specific Demethylase 5D (KDM5D) Isoform Expression in Prostate Cancer Cell Lines: a System Approach

Background: It is now well-demonstrated that histone demethylases play an important role in developmental controls, cell-fate decisions, and a variety of diseases such as cancer. Lysine-specific demethylase 5D (KDM5D) is a male-specific histone demethylase that specifically demethylates di- and tri-methyl H3K4 at the start site of active genes. In this light, the aim of this study was to invest...

Epigenetic regulation of mouse sex determination by the histone demethylase Jmjd1a.

Developmental gene expression is defined through cross-talk between the function of transcription factors and epigenetic status, including histone modification. Although several transcription factors play crucial roles in mammalian sex determination, how epigenetic regulation contributes to this process remains unknown. We observed male-to-female sex reversal in mice lacking the H3K9 demethylas...

Inhibition of histone demethylase JMJD1A improves anti-angiogenic therapy and reduces tumor-associated macrophages.

Antiangiogenic strategies can be effective for cancer therapy, but like all therapies resistance poses a major clinical challenge. Hypoxia and nutrient starvation select for aggressive qualities that may render tumors resistant to antiangiogenic attack. Here, we show that hypoxia and nutrient starvation cooperate to drive tumor aggressiveness through epigenetic regulation of the histone demethy...

The histone demethylase, Jmjd1a, interacts with the myocardin factors to regulate SMC differentiation marker gene expression.

We and others have previously shown that the myocardin transcription factors play critical roles in the regulation of smooth muscle cell (SMC) differentiation marker gene expression. In a yeast 2-hybrid screen for proteins that interact with myocardin-related transcription factor-A (MRTF-A), we identified the histone 3 lysine 9 (H3K9)-specific demethylase, Jmjd1a. GST pull-down assays demonstra...
