P-11: DHR123: A Suitable Fluorescent Mitochondrial Probe for Assessment of H2O2 in Human Spermatozoa

Abstract:

Background: The objective of present study was to assess potential of DHR123 (dihydrorhodamine 123) fluorescent probe for measurement of H2O2 produced by human spermatozoa and comparing the results with DCFH-DA (2&#039;, 7&#039;-dichlorodihydrofluorescein diacetate). Materials and Methods: Fluorescent intensity and percentage R123 and DCF positive sperm measured by flow cytometry. The suitable condition for assessment of H2O2 produced by sperm was 0.05 µM for 1 million sperm per ml for 20 minutes. Results: The results of H2O2 measurement showed a significant correlation (r=0.818, p<0.001) with the DCFH-DA staining routinely used for assessment of H2O2 produced by spermatozoa. H2O2 production was detectable by DHR123 staining in sperm mid-piece. Conclusion: DHR123 is a suitable fluorescent probe for assessment of H2O2 produced by human spermatozoa and can present heterogeneity in H2O2 production between different samples.
