P-11: DHR123: A Suitable Fluorescent Mitochondrial Probe for Assessment of H2O2 in Human Spermatozoa
Authors
Abstract:
Background: The objective of present study was to assess potential of DHR123 (dihydrorhodamine 123) fluorescent probe for measurement of H2O2 produced by human spermatozoa and comparing the results with DCFH-DA (2', 7'-dichlorodihydrofluorescein diacetate). Materials and Methods: Fluorescent intensity and percentage R123 and DCF positive sperm measured by flow cytometry. The suitable condition for assessment of H2O2 produced by sperm was 0.05 µM for 1 million sperm per ml for 20 minutes. Results: The results of H2O2 measurement showed a significant correlation (r=0.818, p<0.001) with the DCFH-DA staining routinely used for assessment of H2O2 produced by spermatozoa. H2O2 production was detectable by DHR123 staining in sperm mid-piece. Conclusion: DHR123 is a suitable fluorescent probe for assessment of H2O2 produced by human spermatozoa and can present heterogeneity in H2O2 production between different samples.

This experiment was designed with the objective of developing a simple, practical, and high repeatability technique for the simultaneous evaluation of the integrity of the plasmatic and acrosomal membranes, as well as funcional mitochondria of domestic fowl spermatozoa using an association of fluorescent probes. Four ejaculates (motility ≥80% and abnormal morphology ≤10%) from each of six Ross ...
full textP-119: In Vitro Assessment of Sucrose for Preservation of Buffalo Epididymal Spermatozoa
Background: Epididymal spermatozoa have been used for production of offspring from genetically important animals after they are dead. To attain optimal fertility with epididymal spermatozoa, they should be preserved in a suitable medium in 37°C. Preservation media provides requirement for spermatozoa and affects its motility, viability, and fertility. Sucrose is one of the important components ...
full textA near-infrared-emitting fluorescent probe for monitoring mitochondrial pH.
We demonstrate a new small molecule fluorescent probe, possessing near-infrared (NIR) emission and an unusually large Stokes shift. It can be readily taken up by live cells and mitochondria, and track subtle pH changes with effectively reduced biological background fluorescence and improved measurement accuracy.
full textDirect nitric oxide measurement in human spermatozoa: flow cytometric analysis using the fluorescent probe, diaminofluorescein.
We have demonstrated that intracellular nitric oxide (NO) can be detected and measured directly in human spermatozoa by flow cytometry using the fluorescent probe 4,5-diaminofluorescein-2/diacetate (10 microM). This method can measure both exogenously added as well as endogenously generated intracellular NO.

Introduction: To evaluate the 8 layer discontinues puresperm gradient in separation of human spermatozoa according to sex chromosomes by fluorescent in situ hybridization (FISH). Material and Methods: This study was carried out on three patient referring to Royan fertility and infertility center. Semen analysis was assessed according to WHO criteria. Each sample was divided into two aliquots, ...
