Abstract:

Background: Thrombopoietin is a glycoprotein produced by liver and kidney which is responsible for regulating the platelet production. Thrombopoietin is a key ligand with impact on regulating the self-renewal of Hematopoietic stem cells and the regulation of Megakaryocytes progenitors. Previous studies have indicated that only N-terminal domain of this protein has receptor promoting ability. The goal of this study was to express the recombinant form of the soluble Thrombopoietin by using Rosetta gami strain of Escherichia coli as the expression host and pET vectors system to produce this cytokine in a cost-effective manner. Methods: After isolating the genomic sequence of Thrombopoietin from HepG2 cDNA, it was inserted into the pET32 vector and colonized by DH5α. Then, recombinant plasmids were transformed into Rosetta gami to express the recombinant protein. Expression was analyzed through SDS-PAGE and western blotting assays. Finally, the optimized time for induction and the suitable concentrations of IPTG were determined.  Results: DNA sequencing of recombinant plasmid proved that the sequence was indicative of variant 2 of Thrombopoietin. SDS-PAGE and western blotting showed a 36-kDa protein band which was compatible with the inserted sequence. Optimized time for expression was 6 h and optimized concentration of IPTG was 0.1 mM. Conclusion: Although different methods are available to produce recombinant Thrombopoietin, low yield and costly setup are major obstacles in the production of Thrombopoietin. Also the products of usual bacterial systems are mostly inclusion bodies which is due to lacking disulfide bond formation. In this study, mutant strain of E. coli was used to provide a high level of water-soluble Thrombopoietin expression.

Gene cloning and expression of soluble thrombopoietin functional domain by harnessing Rosetta-gami expression system

Background: Thrombopoietin (TPO) is an important cytokine that has a critical role in regulating hematopoietic stem cells (HSCs) proliferation and megakaryocyte differentiation. Because of scares amount of this protein in human plasma, in many biotechnological centers around the world, recombinant production of this protein has been carried out. This study was aiming to gene cloning and express...

Soluble Expression and Purification of Q59L Mutant L-asparaginase in the Presence of Chaperones in SHuffle™ T7 strain

Background and Aims: Q59L mutant of L-asparaginase enzyme from Escherichia coli (E. coli) has been introduced with lower side effects. This version of the enzyme might have potential applications in the treatment of leukemia patients. We utilized SHuffle T7 strain of E. coli, to produce the mutant enzyme in the presence of chaperone molecules. Materials and Methods: Q59LAsp gene was cloned in...

Cytotoxicity and Photo-cytotoxcity of Modified, Water- Soluble Carbon Nanotubes in Human Skin Keratinocytes

Water soluble carbon nanotubes have been found to be most promising in biological and biomedical based applications. However, there has been major concern placed on their ability to cause cellular and DNA structural damages upon exposure. Also, influence on natural occurrences such as sunlight, has ignited some apprehension of phototoxicity when exposing to light and carbon based nanomaterials ...

A Novel Vector for Expression/Secretion of Properly Folded Eukaryotic Proteins: a Comparative Study on Cytoplasmic and Periplasmic Expression of Human Epidermal Growth Factor in E. coli

Expression of eukaryotic proteins in E. coli often results in their aggregation. Proper folding and solubility of therapeutical proteins are the pre-requisite for their bioactivity. This is not achieved in cytoplasmic expression in E. coli because of the absence of disulfide bonds formation. A novel expression/secretion vector was constructed which exploited β-lactamase signal sequence to trans...

Expression and Purification of Soluble form of Human Parathyroid Hormone (rhPTH1-34) by Trx Tag in E. coli

Background: Parathyroid Hormone (PTH) is secreted by parathyroid glands and controls the level of calcium in bones and kidney. PTH is a small polypeptide with 84 amino acids, but the first 34 amino acids of which are enough for hormone biological activity and can be used in the treatment of Osteoporosis. The expression efficiency of recombinant human parathyroid hormone rhPTH (1-34) or Teripara...
