Abstract:

Tomato is an important economic crop with nutritional and scientific value for study. Due to its agronomic importance and sensitivity to biotic and abiotic stresses, using new methods in breeding this crop is necessary. In this study, the effects of some factors, such as pre-cultivation time (1, 2, 3 and 4 days), Agrobacterium concentration (OD600nm = 0.4, 0.6, 0.8, and 1), inoculation time (5, 10, 15 and 20 minutes), acetosyringone concentration (0, 100, 150, and 200 μM) and co-cultivation period (1, 2, 3 and 4 days) on the transformation of a local cultivar of tomato called Bonab, were evaluated. The maximum transformation efficiency was obtained at the OD600 = 0.6. Also, the highest transgenic plants were obtained in the 10 minutes of inoculation and 2 days of co-cultivation. The presence of acetosyringone had an increasing effect on the tomato transformation, and the highest percentage of transformation was obtained at 150 M of acetosyringone. Finally, the presence of a chitinase gene in the probable transgenic plants was confirmed by a PCR. The optimal conditions that were obtained in this study can be used to transfer target genes to this tomato cultivar.

The optimization of gene transfer to tomato and the study of expression possibility of salt-tolerance gene (SOS3)

One of the main strategies to improve plant tolerance is the expression of stress-induced genes, which play a significant role in the ionic balance of plants. SOS3 is one of the important components of SOS-regulated ionic homeostasis pathway. Therefore, the expression of this gene could be an important step towards producing salt-resistant plants. In this work, we have transformed tomato (Solan...

Horizontal gene transfer from Agrobacterium to plants

Most genetic engineering of plants uses Agrobacterium mediated transformation to introduce novel gene content. In nature, insertion of T-DNA in the plant genome and its subsequent transfer via sexual reproduction has been shown in several species in the genera Nicotiana and Linaria. In these natural examples of horizontal gene transfer from Agrobacterium to plants, the T-DNA donor is assumed to...

Study of the Factors Affecting Agrobacterium Mediated Gene Transformation in Tomato (lycopersicon Esculentum Mill.) Cv. Riogrande Using Rice Chitinase (cht-3) Gene

The factors affecting the Agrobacterium mediated gene transformation of tomato cv. Riogrande are reported in detail. Shoot tips (explants) were transformed with pBI333 carrying rice chitinase (cht-3) gene. Approximately 49% transformation frequency was achieved. The optimum conditions for successful transformation were: infecting the explants with the Agrobacterium suspension for two minutes, c...

Genetic Manipulation of Tomato (lycopersicon Esculentum) Using Wga Gene through Agrobacterium Mediated Transformation

The major problems faced in the field of Agriculture are loss in crop yield caused by insects, Herbs (weeds), viruses and the pathogens and the pests associated loss is about 14% of total agricultural production. The use of pesticides has resulted in adverse effect on the beneficial organisms and other plant parts such as leaves, fruits and has reached pollution levels, which has become a major...

Agrobacterium-mediated transformation of tomato (Solanum lycopersicum L.) using the expansin 10 (CsEXP10) gene.

The cucumber expansin 10 (CsEXP10) gene was previously cloned from young cucumber fruits but its role has not been defined. To determine the role of this gene in plant growth and development, a CsEXP10 gene transformation system was established. The open reading frame of the gene was inserted behind the CaMV35S promoter of vector pCAMBIA1301, and the construct was introduced into tomato plants ...

Transformation of Cry1a105 gene to tomato in order to increase resistance to Heliothis

Tomato (Solanum lycopersicum L.) is an important vegetable in the world, it exposes to a wide range of pathogens and plant pests attack. Fruit worm is one of the most important the plant pest which mainly damage the fruit and causes the yield reduction. In this study, cry1a105 gene was cloned to pBI121 plasmid (with 35S promoter and Nos terminator), and was transferred to tomato by Agrobacteriu...
