Optimization for high level expression of cold and pH tolerant amylase in a newly isolated Pedobacter sp. through Response Surface Methodology

Authors

Abstract:

Amylase is one of the most widely used enzymes in the industry. Cold environments are the most ubiquitous environments in the world that have been occupied by cold tolerant microorganisms. The enzymes of these microorganisms have a wide range of applications in various areas of biotechnology. The aim of this study was to isolate cold-active amylase producing bacteria. A total of 64 cold-tolerant bacteria producing amylase were isolated from Binaloud Mountain soil, Iran. An isolate (Pedobacter sp. BTR84) registered under accession number KM459538 with the highest enzyme productivity was selected for production optimization. The production of amylase was evaluated via One-factor-at-a-timemethod and RSM (Response Surface Methodology). The enzyme production was optimized at 20°C, pH 9, starch 2% (w/v), and inoculation level 3% (v/v) by One-factor-at-a-timemethod. Then, in order to investigate the interaction between these variables and determine the final optimal conditions, optimization was carried out through the response surface methodology (RSM). Four variables were evaluated at three levels using the Box-Behnken design. Starch concentration and inoculation level variables had a significant effect on amylase production (p

Contemporaneous Production of Amylase and Protease through CCD Response Surface Methodology by Newly Isolated Bacillus megaterium Strain B69

The enormous increase in world population has resulted in generation of million tons of agricultural wastes. Biotechnological process for production of green chemicals, namely, enzymes, provides the best utilization of these otherwise unutilized wastes. The present study elaborates concomitant production of protease and amylase in solid state fermentation (SSF) by a newly isolated Bacillus mega...

Optimization and Modeling of CuOx/OMWNT’s for Catalytic Reduction of Nitrogen Oxides by Response Surface Methodology

A series of copper oxide (CuOx) catalysts supported by oxidized multi-walled carbon nanotubes (OMWNT’s) were prepared by the wet impregnation method for the low temperature (200 °C) selective catalytic reduction of nitrogen oxides (NOx) using NH3 as a reductant agent in the presence of excess oxygen. These catalysts were characterized by FTIR, XRD, SEM-EDS, and H2-TPR meth...

Hydrothermal synthesis of nano-size zirconia using commercial zirconia powder: process optimization through response surface methodology

A hydrothermal method for preparation of nano-size zirconia has been studied to optimize the effective parameters (precursor concentration, temperature and time) using response surface methodology (RSM). The synthesized zirconia samples were characterized through X-ray diffraction (XRD), scanning electron microscope (SEM) and transmission electron microscopy (TEM) analyses to identify mean nano...
