Molecular study of 20-line advanced proteins of durum wheat under water stress

Authors

Abstract:

In order to investigate different lines of durum wheat for water stress tolerance, an experiment in the greenhouse was implemented, using 20 lines of durum wheat at two levels of water stress (40% and 70% of the amount of available water was completely drained in 0.4 and 0.7 levels of stress, respectyively and re-irrigation was performed) and a control level (complete irrigation) in the form of a split plot design in a completely randomized design with three replications. Also, water-soluble and salt-soluble proteins were extracted in the laboratory using SDS-PAGE technique and protein electrophoresis. Measured traits included photosynthetic pigments (chlorophyll a, chlorophyll b, chlorophyll a / b, total chlorophyll and carotenoids), soluble sugar and prolin content. The results of the analysis of variance showed that there was a significant difference between the studied lines at different levels of stress. The results of comparing the mean of the studied traits showed that in total, lines 16 and 5 have the highest value in most of the traits and lines 10 and 18 have the lowest value in most of the traits. Also, the results obtained from the cluster analysis indicated that due to the higher mean values compared to the other lines both in stress-free and stress conditions, lines 16 and 5 have a higher performance stability in the different environmental conditions. Furthermore the results of molecular analysis of the studied proteins showed that in water and salt soluble proteins based on matrix similarity and genetic distance of straw, the lowest genetic distance was between genotypes 18 and 19 (0.32) and the highest genetic distance was between genotypes 16  and 8 (0.98). Finally, it can be concluded that there is a significant genetic diversity between the studied cultivars.

Bound Water in Durum Wheat under Drought Stress.

To study drought stress effects on bound water, adsorption isotherms and pressure-volume curves were constructed for two durum wheat (Triticum durum Desf.) cultivars: Capeiti 8 (drought tolerant) and Creso (drought sensitive). Plants were grown under well-watered and water-stressed conditions in a controlled environment. Differential enthalpy (DeltaH) was calculated through van't Hoff analysis ...

Growth, physiology and yield of durum wheat (Triticum durum) treated with sewage sludge under water stress conditions

In arid and semi-arid areas, low soil fertility and water deficit considerably limit crop production. The use of sewage sludge as an organic amendment could contribute to the improvement of soil fertility and hence the agronomic production. The study aims to highlight the behaviour of durum wheat to the application of sewage sludge associated with water stress. The assessment focused on morphop...

Systems Responses to Progressive Water Stress in Durum Wheat

Durum wheat is susceptible to terminal drought which can greatly decrease grain yield. Breeding to improve crop yield is hampered by inadequate knowledge of how the physiological and metabolic changes caused by drought are related to gene expression. To gain better insight into mechanisms defining resistance to water stress we studied the physiological and transcriptome responses of three durum...

Molecular responses to drought and heat stress in durum wheat

Water scarcity and high temperature stress and their combination are the most important stresses experienced by durum wheat in the ield. The responses induced by drought and heat stress at transcriptional level have been described, but much less is known about plant response to simultaneous drought and heat stress, although this is the most common event in ield conditions. Several data indicate...

Genetic analysis of agronomic and morphological traits of durum wheat [Triticum turgidum L. spp. durum (Desf.)] using generations mean analysis under non-stress and terminal drought stress conditions

Breeders often use generation means analysis to obtain information about inheritance, gene action, and to determine effective breeding methods to improve important agronomic traits, especially under drought stress conditions. To investigate the genetic parameters of agronomic and morphological traits in durum wheat, a field experiment was carried out using F1, F2, BC1 and BC2 generations obtain...

Molecular Docking of Curcumin With Breast Cancer Cell Line Proteins

Background: Breast cancer is known as the most widely recognized dangerous tumors; therefore, the most common reason for mortality among all instances of harmful neoplastic illness in females. This is because the lack of specific signs and symptoms at the early stage and at the aggressive nature. Currently, breast cancer treatment such as chemotherapy, surgery and radiotherapy has not been effe...
