Abstract:

Objective(s) Leishmania can lead to a broad spectrum of diseases, collectively known as leishmaniasis. The A2 gene/ protein family could be one of the most eligible candidate factors of virulence in visceral leishmaniasis (VL). The previous results confirmed that in Leishmania infantum, several A2 proteins are abundantly expressed by the amastigote, but not the promastigote stage. As there are no data available on the pattern of A2 gene / protein in Iranian Leishmania isolates of either cutaneous leishmaniasis (CL) or VL; the current study aimed to investigate molecular analysis of A2 gene in leishmania species among field isolates of Iran. Materials and Methods An A2 gene was identified by sequencing of crude PCR products resulting from 20 samples of CL and 10 samples of VL isolates from Iranian patients. Results The results indicated the A2 gene in CL is only a single copy of 153 bp encoding a protein of 51 amino acids, as opposed to A2 of VL species with multi-copy genes of varying length. A2 sequences in Iranian L. major strains represented a homology with stage-specific S antigen-like protein (A2) of L. major and L. infantum. Moreover, A2 sequences in Iranian L. tropica strains have homology with A2 protein of L. major and L. tropica. Conclusion It is concluded that A2 is an antigen candidate for vaccine development and diagnosis purposes and that A2 sequences are conserved among field isolates.

Genetic Polymorphisms within The Intronless ACTL7A and ACTL7B Genes Encoding Spermatogenesis-Specific Actin-Like Proteins in Japanese Males

Actins play essential roles in cellular morphogenesis. In mice, the t-actin 1 and 2 genes, which encode actin-like proteins, are specifically expressed in haploid germ cells. Both T-ACTIN 1/ACTLB and T-ACTIN 2/ACTL7A have also been cloned as orthologous genes in humans; they are present on chromosome 9q31.3 as intronless genes. Defects of germ cell-specific genes can introduce infertility witho...

Leishmania major-like antigen for specific and sensitive serodiagnosis of human and canine visceral leishmaniasis.

An antigen (LMS) prepared from Leishmania major-like promastigotes was used in an enzyme-linked immunosorbent assay (ELISA) for the diagnosis of human and dog visceral leishmaniasis. The results were compared with those from the indirect immunofluorescent antibody test (IFAT). A total of 1822 canine sera were tested, including sera from dogs with visceral leishmaniasis, transmissible venereal t...

isoenzyme characterization of iranian leishmania isolates from cutaneous leishmaniasis

during the period from 1996 to 1997, 45 isolates of leishmania were recovered frompatients suspected of cutaneous leishmaniasis (cl) and one from a gerbil. these specimens werereceived from various parts of iran. isoenzyme profiles of these isolates were compared with those ofreference strains of l.tropica, l.major and l.infantum using cellulose acetate electrophoresis (cae) and10 enzyme system...

Analysis of Plasmodium vivax Apical Membrane Antigen-1 (PvAMA-1) Haplotypes among Iranian Isolates

Plasmodium vivax apical membrane antigen-1(PvAMA-1) is a surface protein with polymorphic sites. This study was aimed to analyze the polymorphic amino acid residues at PvAMA-1 in different infected age groups. 92 blood samples were collected from south and southeast of Iran. The DNA coding for the domain I (DI), DII, and partial DIII of this antigen was amplified by Nested-PCR, and sequenced. N...
