Abstract:

Objective(s) Nicotinic acetylcholine receptors (nAChRs) regulate epileptiform activity and produce a sustained pro-epileptogenic action within the hippocampal slices. In the present study, we investigated the effect of nAChRs on evoked glutamatergic synaptic transmission in area CA3 and CA1 of rat hippocampal slices to identify possible excitatory circuits through which activation of nAChRs produce their pro-epileptogenic effects. Materials and Methods   Hippocampal slices (400 µm thick) prepared in vitro from male Wistar rats (3-5 weeks), using standard procedures. Following 1 hr equilibration in artificial cerebrospinal fluid (ACSF), slices transferred to an interface recording chamber. Stimulatory electrodes placed within the hilus or Schaffer-collateral pathways and extracellular field recordings made in the stratum radiatum of the CA1 and CA3 regions to investigate evoked synaptic responses. Results Bath application of the selective nAChR agonist dimethylphenyl-piperanzinium (DMPP, 30 mM) resulted in a sustained and reversible enhancement of glutamate afferent evoked fEPSP amplitude by 15.7±5.1%            (mean±SEM; n=8 of 12) in the CA3 region of the hippocampus but not in the CA1 (-5.25±8.3%, mean±SEM; n=5). Conclusion Activation of nAChRs may produce pro-epileptogenic actions in part through regulating glutamatergic circuits. Difference in nAChR regulation is also evident between different regions of hippocampus.

Modulation of nicotinic acetylcholine receptors by strychnine.

Strychnine, a potent and selective antagonist at glycine receptors, was found to inhibit muscle (alpha1beta1gammadelta, alpha1beta1gamma, and alpha1beta1delta) and neuronal (alpha2beta2 and alpha2beta4) nicotinic acetylcholine receptors (AcChoRs) expressed in Xenopus oocytes. Strychnine alone (up to 500 microM) did not elicit membrane currents in oocytes expressing AcChoRs, but, when applied be...

The modulation of nicotinic acetylcholine receptors on the neuronal network oscillations in rat hippocampal CA3 area

γ oscillations are associated with higher brain functions such as memory, perception and consciousness. Disruption of γ oscillations occur in various neuro-psychological disorders such as schizophrenia. Nicotinic acetylcholine receptors (nAChR) are highly expressed in the hippocampus, however, little is known about the role on hippocampal persistent γ oscillation. This study examined the effect...

Indirect modulation by alpha7 nicotinic acetylcholine receptors of noradrenaline release in rat hippocampal slices: interaction with glutamate and GABA systems and effect of nicotine withdrawal.

Nicotinic acetylcholine receptors (nAChRs) can modulate transmitter release. Striatal [(3)H]dopamine ([(3)H]DA) release is regulated by presynaptic nAChR on dopaminergic terminals and alpha7 nAChR on neighboring glutamatergic afferents. Here, we explored the role of alpha7 nAChR in the modulation of [(3)H]noradrenaline ([(3)H]NA) release from rat hippocampal slices. The nicotinic agonist anatox...

Deficits of synaptic functions in hippocampal slices prepared from aged mice null α7 nicotinic acetylcholine receptors.

Alpha 7 (α7) nicotinic acetylcholine receptor (α7-nAChR) is one of most high expressed nAChR subtypes in the brain. The activation of nAChRs enhances animal cognitive, learning and memory abilities. However, the role of genetic knockout (KO) of α7-nAChRs in animal cognition-associated behaviors is still obscure. An early report showed that α7-nAChR KO mice did not exhibit behavioral phenotypes,...
