kidd blood group genotyping in alloimmunized thallasemia patients
Authors
Abstract:
Abstract Background and Objectives Hemagglutination has limitations in identifying the phenotype of patients who have been recently transfused due to the presence of donor red cells (RBCs) in the patient’s circulation. Kidd blood group is one of the most important blood groups in transfusion medicine and related antibodies are responsible for one third of delayed haemolytic transfusion reactions (DHTRs). In this study we developed a molecular procedure for detection of alleles responsible for Kidd blood group in thalassemia patients. Materials and Methods In this cross sectional descriptive study, one hundred of alloimmunized thalassemic patients from Tehran Adult Thalassemic Clinic in 2017 were studied. The phenotype of samples was tested by routine serological methods. PCR-SSP, PCR-RFLP and DNA sequencing were used to determine the genotype of kidd blood group for these patients. Results Discrepancies were found between the phenotype and genotype in 20 out of 100 cases. In 11(55%) cases phenotype was determined as Jk (a+b+) but genotype was JKA/JKA, in 7 (35%) cases phenotype was Jk (a+b+) while the genotype showed JKB/JKB, 1(5%) case had been phenotyped as Jk (a+b-) but it was genotyped as JKA/JKB, and 1(5%) case had been phenotyped as Jk (a-b+) but it was genotyped as JKA/JKB. Conclusions Molecular techniques as a complementary method together with the results of serological methods can help resolve the problems caused by blood transfusion and bring about accurate determination of phenotype in thalassemic patients with multiple blood transfusions.

Rh blood group genotyping in alloimmunized thalassemic patientsin Tehran Adult Thalassemia Clinic during 2017-18
Abstract Background and Objectives The main antigens of the Rh blood group system (D,C,c,E,e) after the ABO system, are the most immune antigens. Due to existence of donor's red blood cells in the patient's circulation serologic methods can not accurately detect blood group antigens in patients with chronic blood transfusions, but molecular methods can overcome many of these limitations. ...

BACKGROUND Molecular testing is more precise compared to serology and has been widely used in genotyping blood group antigens. Single nucleotide polymorphisms (SNPs) of blood group antigens can be determined by the polymerase chain reaction with sequence specific priming (PCR-SSP) assay. Commercial high-throughput platforms can be expensive and are not approved in China. The genotype frequencie...
full textBlood Group ABO Genotyping in Paternity Testing.
BACKGROUND: The ABO blood groups result from DNA sequence variations, predominantly single nucleotide and insertion/deletion polymorphisms (SNPs and indels), in the ABO gene encoding a glycosyltransferase. The ABO blood groups A(1), A(2), B and O predominantly result from the wild type allele A1 and the major gene variants that are characterized by four diallelic markers (261G>del, 802G>A, 803G...
full textPrevalence of Rh, Duffy, Kell, Kidd & MNSs blood group antigens in the Indian blood donor population
BACKGROUND & OBJECTIVES Little data are available regarding the frequencies of the blood group antigens other than ABO and RhD in the Indian population. Knowledge of the antigen frequencies is important to assess risk of antibody formation and to guide the probability of finding antigen-negative donor blood, which is especially useful when blood is required for a patient who has multiple red ce...

BACKGROUND Duffy (FY) blood group genotyping is important in transfusion medicine because Duffy alloantibodies are associated with delayed hemolytic transfusion reactions and hemolytic disease of the fetus and newborn. In this study, FY allele frequencies in Thai blood donors were determined by in-house PCR with sequence-specific primers (PCR-SSP), and the probability of obtaining compatible bl...

OBJECTIVE Conventional serologic typing of red blood cell systems other than ABO and RhD can be inaccurate and difficult to interpret in patients who have recently undergone blood transfusion. While molecular-based assays are not used routinely, the usefulness of genotyping was investigated in order to determine patients who may benefit from this procedure. MATERIALS AND METHODS Blood samples...
