Abstract:

Background and Aims: In the current study, two bacteriophages were isolated from the wastewater sources and characterized by lytic activity against the Listeria monocytogenes (L. monocytogenes) bacterium. Materials and Methods:  50 wastewater samples were collected from the disposal sites. The phages were isolated from the treated samples using the double agar overlay method with L. monocytogenes as the host bacterium. Plaque morphology, temperature, pH, titration of phage lysate, host range, and scanning electron microscopic morphology were used to characterize the isolated phages. Results: Bacteriophage was found in two samples taken from wastewater treatment plants. Phages were proliferated and released under optimal bacterial growth conditions at pH 7; 37 °C in 36-hour incubation. A 10:1 ratio of bacterial cells to phage produced the greatest lytic ability. The highest phage titrated under optimal conditions was estimated at 9×109 pfu/ml. The scanning electron microscopic image revealed a bacteriophage morphology corresponding to the head and tail group. The host range observations demonstrated that L. monocytogenes were unique to the isolated phage. The elimination of L. monocytogenes growth in milk and whey samples revealed that this bacteriophage targeted bacteria and resulted in a 75% reduction in bacterial contamination. Conclusion: The results suggest that the phages obtained from L. monocytogenes may be utilized to combat this pathogen in dairy, agri-food, wastes, and other associated infections.

Isolation of Listeria monocytogenes from Meat and Dairy Products

Introduction: This study was intended to determine the presence and distribution of Listeria monocytogenes in various meat and dairy products from Qazvin Province by culture followed by biochemical and morphological assays. The identity of the isolates was further obtained by amplification of prfA gene in bacteria isolates. This gene is a transcriptional activator of virulence gene ex...

Isolation of lytic bacteriophages against pathogenic Escherichia coli strains in poultry in the northwest of Iran

In this study, 90 internal organ samples of poultry with symptoms of colibacillosis were obtained from Maragheh poultry farms in East Azerbaijan, Iran. In total, 70 bacterial isolates were confirmed as Escherichia coli (E. coli) strains using standard biochemical tests, and antibiotic sensitivity was determined by the disk diffusion method. Antibiotics used in this study included ampicillin, pe...

Biocontrol of Listeria monocytogenes on fresh-cut produce by treatment with lytic bacteriophages and a bacteriocin.

The fresh-cut produce industry has been the fastest-growing portion of the food retail market during the past 10 years, providing consumers with convenient and nutritious food. However, fresh-cut fruits and vegetables raise food safety concerns, because exposed tissue may be colonized more easily by pathogenic bacteria than intact produce. This is due to the higher availability of nutrients on ...

Isolation and characterization of bacteriophages with lytic activity against common bacterial pathogens

AIM Present investigation was conducted to isolate and characterize bacteriophages with lytic activity against common bacterial pathogens. MATERIALS AND METHODS A total of 60 samples of animal waste disposal from cattle (42) and buffalo (18) farms were collected from three different strata, i.e., top, mid, and bottom of collection tank. Samples were primarily subjected to rapid detection meth...

Isolation of Listeria monocytogenes from raw milk.

During a recent outbreak of listeriosis, we examined 121 raw milk samples and 14 milk socks (filters). Listeria monocytogenes was recovered from 15 (12%) of 121 milk specimens and 2 (14%) of 14 milk socks. The optimal processing method consisted of cold enriching diluted milk for 1 month with culture to selective broth, followed by plating.

Characterization of Newly Isolated Lytic Bacteriophages Active against Acinetobacter baumannii

Based on genotyping and host range, two newly isolated lytic bacteriophages, myovirus vB_AbaM_Acibel004 and podovirus vB_AbaP_Acibel007, active against Acinetobacter baumannii clinical strains, were selected from a new phage library for further characterization. The complete genomes of the two phages were analyzed. Both phages are characterized by broad host range and essential features of pote...
