Abstract:

Di(2-ethylhexyl)phthalate (DEHP) is a plasticizer in a variety of consumer products. Given the adverse toxicity effects of DEHP on living organisms, its high consumption and dispersal has been caused a serious concern. . Nowedays, the Fish Embryo Test (FET) method is widely used among the conventional methods of toxicology .The aim of the this study was to determine the acute toxicity of Di(2-ethylhexyl)phthalate on zebrafish (Danio rerio) embryo using FET method to assess both lethal and sublethal endpoints and anomalies effects over 5 days . Zebrafish embryos (3 hours post-fertilization) were exposed to different concentrations of DEHP (0.05, 5, 20, 35, 50 and 100 µg/l) for 120 h, then, lethal and sub-letahl endpoints of embryos evaluated and data were analized using Probit software. The LC50, NOEC and LOEC values obtained for DEHP were 46.14, 12.08 and 22.06 µg/l, respectively. Also, There were abnormalities including  coagulation of the embryo, lack of somite formation, non-detachment of the tail bud from the yolk sac, malformation of tail and lack of heartbeat in the embryos. This study showed that DEHP is a teratogenic pollutants and the application of FET method is / an alternative method for developmental toxicity tests to predict effects of emerging pollutants on aquatic organisms.

Evaluation of the Toxic Effects of Clozapine in Zebra fish (Danio rerio) embryos with the Fish Embryo Toxicity Test

Clozapine (an atypical antipsychotic drug) is indicated in treatment-refractory schizophrenia. Antipsychotics have been reported to have cardiotoxic effects in humans. The zebra fish (Danio rerio) is a tropical cyprinid used in toxicological research. In the present study, the toxicity of clozapine was evaluated in zebra fish embryos with the aid of the fish embryo toxicity test (FET). The endpo...

Evaluation of of the performance of the zebrafish (Danio rerio) model in nanotoxicology studies with emphasis on embryo pathology

The present study was conducted to evaluate the performance of zebrafish (Danio rerio) as a model in embryo and fetal pathology in nanotoxicology studies. Examination of the sources showed that it is possible to completely inhibit hatching and fetal death when exposed to nanoparticles, because nanoparticles interact with hatching enzymes. Zebrafish embryo developmental abnormalities have been s...

DarT: The embryo test with the Zebrafish Danio rerio--a general model in ecotoxicology and toxicology.

The acute fish test is an animal test whose ecotoxicological relevance is worthy of discussion. The primary aim of protection in ecotoxicology is the population and not the individual. Furthermore the concentration of pollutants in the environment is normally not in the lethal range. Therefore the acute fish test covers solely the situation after chemical spills. Nevertheless, acute fish toxici...

Danio Rerio, the Zebrafish

The zebrafish has long been a favourite model for the study of vertebrate development. Here we provide an overview of the current state of knowledge and resources for the study of this fish, with comments on the future direction of zebrafish genomics from Professor Mark Fishman and Dr Stephen Wilson.

BIOLOGY OF ZEBRAFISH, Danio rerio

Zebrafish, Danio rerio (Hamilton), is one of the most important vertebrate model organism in genetics, developmental biology, neurophysiology and biomedicine (Rubinstein, 2003; Amsterdam and Hopkins, 2006). It is a small, robust fish, so large numbers can be kept easily and cheaply in the laboratory, where it breeds all year round. Development is rapid, with precursors to all major organs devel...

Effects of stanniocalcin 1 on calcium uptake in zebrafish (Danio rerio) embryo.

Stanniocalcin (STC) formerly called hypocalcin or teleocalcin, is a 50-kDa disulfide-linked homodimeric glycoprotein that was originally identified in fish and secreted from the corpuscles of Stannius (CS). One of the main functions of STC-1 is Ca(2+) uptake inhibition; however, the mechanisms remain unknown. In the present study, we provide molecular evidence to elucidate how zebrafish STC-1 r...
